A new method for fast isolation of insect antifeedant compounds from complex mixtures

Abstract: Bioassay-guided isolation of bioactive natural substances requires monitoring of all fractionation and purification steps using a bioassay system. This is often a long and tedious process, especially with insect feeding bioassays. We report here a new method, based on the principle of bioautography, for a quick isolation of insect antifeedant compounds. TLC plates, after development, are coated with a thin layer of artificial diet and fed toSpodoptera litura larvae. The location of uneaten areas is then compar… Show more

“…Skimmia japonica has been screened for antifeedants using Spodoptera exigua, but only furanocoumarins were reported to be responsible for the deterrent properties of the plant (Escoubas et al, 1992). Our results indicate that furoquinolines also contribute to these effects.…”

“…It has to be pointed out that the obtained results induce to the occurrence of synergic effect between the substances, so that each single compound or compound class presents bioactivity, which is magnified when the compounds are present in the same sample. Synergic effects between terpenes and terpenoids have already been reported by Escoubas et al 48 and Iscan et al, 49 among other authors.…”

“…Our method presented here is not unique in its use of TLC plates (Metcalf et al 1980;Escoubas et al 1992) or other artificial substrates for Coleoptera (Nawrot et al 1991;Hare 1998). However, the short test period and the low demand of test chemicals make it a microbioassay, in line with the trend toward downsizing .chemical methods in chemical ecology.…”

“…Metcalf et al (1980) described feeding stimulation by direct testing of curcurbitacins separated on TLC (thin layer chromatography) plates. Escoubas et al (1992) described a feeding assay for insect larvae, including direct testing of chromatographic separations on TLC plates for antifeedant compounds.…”
