A natural process of cirrhosis resolution and deceleration of liver regeneration after thioacetamide withdrawal in a rat model

Abstract: Characteristics of thioacetamide (TAA)-induced liver cirrhosis in rat was observed for 120 days after TAA withdrawal as part of the radiobiological study of partial liver irradiation on TAA-induced cirrhotic rats. The natural process focused on cirrhosis and regeneration was recorded as a baseline condition for the interpretation of the outcome of the partial liver irradiation study. Cirrhosis in rats was successfully induced by drinking 0.03% TAA water orally for 29 weeks with a modeling rate of 96%. After es… Show more

“…Immunohistochemical staining was performed as described previously [42]. The expression levels of cyclin B1, Sec62, and Birc3 were calculated by the number of positive cells per 1000 hepatocytes counted, which was defined as LI.…”

“…Sections (five microscopic fields per slice with one slice/animal) were digitized using an Olympus BX51 microscope (Olympus Optical Co, Ltd, Tokyo, Japan). Digital images were processed using Image-Pro Plus 6.0 software (Media Cybernetics Inc, Maryland, USA), which was used to determine the integral optical density (IOD) and average optical density (AOD, AOD=positive area×OD/total area) of p22 phox , p47 phox , and eNOS as described previously [34,35] .…”

“…In the present study, we used rats with normal liver function as recipients. It could be speculated that the proliferation and differentiation of hepatic progenitor cells may be activated by partial hepatectomy in rats which already have a dysfunctional liver, as would be the case in cirrhosis, because these livers have defects in regeneration due to their pre‐existing damage (Gu et al ., ).…”
