A Nanoscale Metal–Organic Framework to Mediate Photodynamic Therapy and Deliver CpG Oligodeoxynucleotides to Enhance Antigen Presentation and Cancer Immunotherapy

Abstract: Checkpoint blockade immunotherapy (CBI) awakes a host innate immune system and reactivates cytotoxic T cells to elicit durable response in some cancer patients. Now, a cationic nanoscale metal–organic framework, W‐TBP, is used to facilitate tumor antigen presentation by enabling immunogenic photodynamic therapy (PDT) and promoting the maturation of dendritic cells (DCs). Comprised of dinuclear WVI secondary building units and photosensitizing 5,10,15,20‐tetra(p‐benzoato)porphyrin (TBP) ligands, cationic W‐TBP … Show more

“…Through solvothermal reactions between benzoporphyrin and WCl 6 , benzoporphyrin was incorporated into a W-based nMOF [72]. Anionic CpG as an adjuvant was adsorbed on the surface of the cationic nMOF.…”

“…(4) Relatively labile coordination bond connections make nMOFs intrinsically biodegradable in physiological environments for biocompatibility. As a result, nMOFs have been successfully applied to the delivery of chemotherapeutic agents, [200][201][202] imaging contrast agents, 172, 203 photosensitizers, [204][205][206][207][208][209][210][211][212][213] proteins [214][215] and gene therapy drugs. [216][217][218] In this dissertation, I report the progress on the design of nMOFs as nanosensitizers to enhance RT, 219 mediate radiotherapy-radiodynamic therapy (RT-RDT), [220][221] and enable PDT and CDT 222 with the advantages of high payload, crystalline order, and porous structures for ROS generation.…”

“…Then the immunotherapy agent called IDO, which could inhibit the express of indoleamine 2,3‐dioxygenase, was encapsulated into the channels of NMOFs, so as to achieve the synergistic effect of PDT and immunotherapy for inhabiting colorectal cancers. In addition to PDT/immunotherapy, some other therapies have also been combined with the NMOFs mediated PDT, especially chemotherapy, owing to that chemotherapeutic drugs could easily be encapsulated into the pores of NMOFs. Various experimental results turned out that the synergistic effect was better than the single therapy.…”
