A molecular linkage map of cultivated oat

Abstract: A molecular linkage map of cultivated oat composed of 561 loci has been developed using 71 recombinant inbred lines from a cross between Avena byzantina cv. Kanota and A. sativa cv. Ogle. The loci are mainly restriction fragment length polymorphisms detected by oat cDNA clones from leaf, endosperm, and root tissue, as well as by barley leaf cDNA clones. The loci form 38 linkage groups ranging in size from 0.0 to 122.1 cM (mean, 39 cM) and consist of 2-51 loci each (mean, 14). Twenty-nine loci remain unlinked. … Show more

“…K Pellizzaro et al population encompassed a total genetic distance of 1,397.5 cM (Figure 1). These results are in agreement with the results of molecular mapping previously performed in hexaploid oats (O'Donoughue et al 1995, Wight et al 2003, Zhu and Kaeppler 2003, Tinker et al 2009). The expected number of linkage groups is 21, corresponding to the number of chromosomes in each haploid cell (n) of hexaploid oat.…”

“…After this procedure, the expected size-restriction fragments were detected when probing the Southern blots with total lambda DNA. Additional confirmation of complete sample digestion was obtained by probing Southern blots with an oat restriction fragment length polymorphism (RFLP) probe CDO 638 (39), which consistently detected the same restriction fragments (ranging from 6 to 23 kb in size) in all samples treated with the same restriction endonuclease (data not shown). All 13 single-locus lines exhibited multiple DNA fragments that hybridized to the mixed probe consisting of all transformed plasmids (Fig.…”

“…'Kanota' x A. sativa L. cv. 'Ogle' (K x O) cobria uma distância de 1,482,0cM (O'DONOUGHUE et al, 1995). Já o mapa desenvolvido por ZHU & KAEPPLER (2003), para uma população oriunda do cruzamento entre as cultivares Ogle x MAM17-5, cobre uma extensão de 1.396,7cM e possui 28 grupos de ligação.…”
