A mimotope of Pre-S2 region of surface antigen of viral hepatitis B screened by phage display

Abstract: To acquire the phage-displayed mimotopes which mimic the specificity of hepatitis B virus surface antigen (HBsAg), a random peptide library expressing linear peptide with 12 amino acids in length were used to screen with the serum from a hepatitis B virus infected patient in the recovery phase. After 3 rounds of biopanning, the positive phages were confirmed by competitive ELISA using HBsAg/P33. Two phagotopes were identified and one of them was confirmed as mimotope by competition experiment. Based on the mim… Show more

“…Recent work has shown that phage-displayed peptides selected using antibodies raised against pathological antigens can be an important tool for both diagnosis and disease prevention (Delmastro et al, 1997;Enshell-Seiffers et al, 2001;Irving et al, 2001;Zhang et al, 2001;Marston et al, 2002;Beckman et al, 2005). Such phagedisplayed peptides (mimotopes) do not necessarily have sequence homology with the antigen, but have sufficient conformational homology to induce high affinity antibodies that bind to both the mimotope and the natural antigen (Collins, 1997).…”

“…In most cases, peptide phage display libraries displaying 7–15mer random peptides are screened on immobilised antibodies to identify immungenic epitopes. This results only in the identification of short mimotopes (sequences mimicking the epitope) [82,83,84]. Also the identification of conformational epitopes is hampered by the short peptides displayed by random peptide libraries.…”

“…A random peptide library has been used to acquire the phage-displayed mimotopes that mimic the specificity of HBsAg. Two phagotopes have been identified and one of them was confirmed as a mimotope by competition experiment [160]. Octavalent MAP (MAP 1) containing the mimotope TANGFYRLPSGS and control MAP (MAP 2) based on the epitope of AAs 130-150 of HBsAg have been prepared by SPPS.…”
