Several animal and clinical studies have shown that phytoestrogens, plant-derived estrogenic compounds, can be useful in treating postmenopausal osteoporosis. Phytoestrogens and phytoestrogen-containing plants are currently under active investigation for their role in estrogen-related disorders. The present study deals with anti-osteoporotic evaluation of phytoestrogen-rich plant Cuminum cyminum, commonly known as cumin. Adult Sprague-Dawley rats were bilaterally ovariectomized (OVX) and randomly assigned to 3 groups (10 rats/group). Additional 10 animals were sham operated. OVX and sham control groups were orally administered with vehicle while the other two OVX groups were administered 0.15 mg/kg estradiol and 1 g/kg of methanolic extract of Cuminum cyminum fruits (MCC) in two divided doses for 10 weeks. At the end of the study blood, bones and uteri of the animals were collected. Serum was evaluated for calcium, phosphorus, alkaline phosphatase and tartarate resistant acid phosphatase. Bone density, ash density, mineral content and mechanical strength of bones were evaluated. Scanning electron microscopic (SEM) analysis of bones (tibia) was performed. Results were analyzed using ANOVA and Tukeys multiple comparison test. MCC (1 g/kg, p.o.) significantly reduced urinary calcium excretion and significantly increased calcium content and mechanical strength of bones in comparison to OVX control. It showed greater bone and ash densities and improved microarchitecture of bones in SEM analysis. Unlike estradiol it did not affect body weight gain and weight of atrophic uterus in OVX animals. MCC prevented ovariectomy-induced bone loss in rats with no anabolic effect on atrophic uterus. The osteoprotective effect was comparable with estradiol.

Several causative factors have been proposed for the occurrence of skeletal anomalies in fish larvae, among which we quote nutritional factors, such as LC-PUFAs. This study aimed to investigate the effect of different dietary DHA and ARA level and ratio on pikeperch (Sander lucioperca) larval development and performance, digestive capacity, fatty acids composition, skeleton anomalies and molecular markers of oxidative stress status (sod, gpx, and cat), stress response (StAR, gr, pparα, hsl and pepck), fatty acid synthesis (fadsd6, elovl5), eicosanoids synthesis (pla2, cox2, lox5, pge2, and lta4h), and bone development (twist, mef2c, sox9, and alp). Pikeperch larvae were fed six microdiets containing two different dietary levels of DHA (0.5% and 3.5%) combined with three levels of ARA (1.2%, 0.6%, and 0.3%). Dietary fatty acid changes did not affect growth performance but significantly influenced enzymatic activities. A significant increase in skeletal anomalies with DHA intake increment was recorded. StAR, cox2, pla2 and hsl expression were significantly depressed in 2.5% DHA larvae. An opposite effect of dietary DHA elevation was recorded in gpx expression. Both DHA and ARA had a significant effect on pparα, gr, and pge2 expressions. Although no significant interactions were found, pge2, gr, and pparα displayed a differential pattern of expression between the different treatments. A strong association was found for the larval tissue amount of ARA and DHA with eicosanoid metabolism, stress response and skeleton anomaly related genes. These results denoted the effects of dietary LC-PUFAs on immune/stress gene regulation and their potential implication in skeleton development.

The present work aimed to study some blood indices of rats as affected by saccharin and the therapeutic action of vitamins C and E. The used adult female Rattus norvegicus albino rats in the present study were weighing 100—120 g. Administration of saccharin at a dose of 35 mg kg—1 body weight (b.wt.) day—1 for 35 days significantly decreased serum glucose, triglycerides, cholesterol, total protein and albumin values. These decrements were by 20.16%, 22.76%, 44.92%, 20.16% and 40.44%, respectively, compared to control level (p value < 0.01). But it increased levels of kidney function indices. The effect of saccharin was more pronounced on creatinine. Activities of Alanine aminotranferease (ALT), aspartate aminotransferase (AST) and Alkaline phosphatase (ALP) increased significantly following saccharin treatment to rats. Concerning hematoligical parameters, the more obvious changes were observed in the increment of white blood cell (WBC), mean corpuscular volume (MCV) and platelets (PLT) and the decrease in hematocrit, hemoglobin (Hb) and red blood cells (RBCs) count in response to the administration of saccharin. In general, vitamin C or E (150 mg kg—1 b.wt. day—1 for 35 days) was able to reduce the effects of saccharin intake. Both vitamins, however, generally have beneficial effects in reducing the changes in the studied parameters.
