A Mannosylated, PEGylated Albumin as a Drug Delivery System for the Treatment of Cancer Stroma Cells

Abstract: Mannose receptors that are expressed on macrophages and fibroblasts in cancer stroma are promising therapeutic targets for cancer treatment. Albumin can be used as a drug carrier in chemotherapeutics due to its accumulation in the tumor tissue by the enhanced permeability and retention effects. A mannosylated albumin was recently developed as a new drug carrier targeting cells that express mannose receptors such as macrophages and fibroblasts in cancer stroma. The mannosylated albumin is specifically distribut… Show more

“…Man-BSA was first synthesized through the Schiff base reaction between CHO-Man and NH2-BSA ( Figure 1A ; Mizuta et al, 2021 ). The structure of Man-BSA was demonstrated by 1 H-NMR, which was reflected in the characteristic absorption peak of 0.5–3.0 ppm (Man) and 3.2–3.9 ppm (BSA) ( Figure 1B ).…”

“…Dual-modified albumin which was reported by Mizuta et al, incorporating mannosylation and polyethylene glycosylation, enhances blood circulation and stromal cell targeting, facilitating efficient paclitaxel delivery and ensuing tumor disruption (Figure 6B). 76 Chemotherapy, while efficacious against primary tumors, can paradoxically promote metastasis through the release of proinflammatory factors. Li et al developed novel polymeric nanoparticles that adeptly deliver chemotherapeutics and scavenge proinflammatory factors, curbing chemotherapyinduced breast cancer metastasis.…”

“…The patented nanoparticle albumin-bound (NAB) technology, which consists of the evaporation of an emulsion with the creation of cross-links between albumin units, is the best-known preparation method for albumin-based nanoparticles [105,106]. In addition to Abraxane ® , which is created with the help of NAB technology and has been successfully used in clinical practice [111][112][113], work is also underway to create a range of albumin-based nanoparticles [104][105][106][107][108][109][110][111][112][113][114][115][116][117][118].…”
