A malaria vaccine adjuvant based on recombinant antigen binding to liposomes

Abstract: Pfs25 is a malaria transmission-blocking vaccine antigen candidate, but its apparently limited immunogenicity in humans has hindered clinical development. Here, we show that recombinant, his-tagged Pfs25 can be mixed at the time of immunization with pre-formed liposomes containing cobalt-porphyrin-phospholipid (CoPoP), resulting in spontaneous nanoliposome antigen particleization (SNAP). Antigens are stably presented in uniformly-oriented display via his-tag insertion in the CoPoP bilayer, without covalent mod… Show more

“…The same group used liposomes modified with a fragment of tetanus toxoid as an antigen and PEGylated peptide adjuvants to target the Fc receptor of dendritic cells as a cancer vaccine . Recently, Lovell and co‐workers appended a recombinant polyhistidine‐tagged version of a malaria transmission‐blocking antigen (Pfs25) to the surfaces of liposomes, which induced a potent humoral immune response mediated by dendritic cell cross‐presentation …”

“…The previous examples discussed the importance of antigen density in the context of autoimmune disease, but this feature is also important in mounting immune responses against infectious diseases. As one illustration, recent studies have exploited liposomes to control how malaria antigens are presented . These liposomes are designed to load antigens into the membrane stably by coordinating interactions with metal ions located in the particles.…”
