A Magnetic Bead-Based Sensor for the Quantification of Multiple Prostate Cancer Biomarkers

Abstract: Novel biomarker assays and upgraded analytical tools are urgently needed to accurately discriminate benign prostatic hypertrophy (BPH) from prostate cancer (CaP). To address this unmet clinical need, we report a piezeoelectric/magnetic bead-based assay to quantitate prostate specific antigen (PSA; free and total), prostatic acid phosphatase, carbonic anhydrase 1 (CA1), osteonectin, IL-6 soluble receptor (IL-6sr), and spondin-2. We used the sensor to measure these seven proteins in serum samples from 120 benign… Show more

“…The capability of applying precise intracellular forces circumvents the cytoskeleton and directly exerts forces on the cell nucleus. Besides biophysical measurement, the submicrometer magnetic bead can also be chemically functionalized (e.g., with pH-sensitive or Ca 2+ -sensitive fluorophores) for intracellular biochemical sensing (62,63).…”

“…Another benefit of this detection method is the potential to detect antigens in the gas phase as well as in the liquid phase. A piezoelectric membrane-based approach with the application of magnetic beads to quantify seven PCa biomarkers including PSA was described [146]. The study dealt with the discrimination between 100 PCa (Gleason score 6 and 7 – a grading system used to evaluate the prostate biopsy samples) and 120 BPH patients.…”

“…The study dealt with the discrimination between 100 PCa (Gleason score 6 and 7 – a grading system used to evaluate the prostate biopsy samples) and 120 BPH patients. For these seven proteins in serum samples, the highest AUC (area under the curve) discrimination was achieved with a spondin-2 or free/total PSA operation where the area under the curve was 0.84 with a p value below 10 −6 [146].…”
