A light-induced nitric oxide controllable release nano-platform based on diketopyrrolopyrrole derivatives for pH-responsive photodynamic/photothermal synergistic cancer therapy

Abstract: An intelligent multifunctional nano-platform responsive to the tumor microenvironment was established, which showed NO controllable “on–off” release and enhanced photodynamic/photothermal synergistic cancer therapy.

“…Figure d shows that this logic device can respond well to ATP at a concentration of over 1 m m , which is compatible with cellular ATP level (1–10 m m ) . Likewise, the device functions well when lowering pH to below 5 (Figure e), near the lysosomal pH range (approximately 4.7) . These observations suggest that this FNA logic device would function well in living cells.…”

“…Such a size could promote better aggregation of the nanomedicine at a tumor site via an EPR effect. [65][66][67] As displayed in Fig. S1, † the particle size of UCNPs@mCeO x changed from 82 nm to 103 nm owing to the polymer coating, the etching of the mesoporous silica shell and the adsorbed water molecules on the NP surface.…”

“…Three peaks with similar intensity show the existence of TEMPO, which manifests the generation of 1 O 2 . Since the major generation of •OH radicals depended on the electrochemically assisted Fenton-like reaction by the active center of Fe 3+ and Mn 2+/3+ through the single electron transfer (Equations (2), (5), and (6)), the detection of •OH in the H 2 O 2 solution without laser radiation is essential. The free •OH can be captured by spin traps 5,5-dimethyl-1-pyrroline-Noxide (DMPO, 0.02 m) to obtain the DMPO-OH spin adduct, which has a strong ESR spectra.…”

“…To verify the theory qualitatively, 3,3′,5,5′-tetramethyl-benzidine (TMB) was applied to monitor the radical production. The steady-state catalytic kinetics of FeMn-LDH were performed in an acidic TMB solution (pH 6.5) using different concentration H 2 O 2 (5,10,20, and 40 × 10 −6 m) as substrates in acid phosphate buffer saline (PBS), respectively. Then, the initial reaction rates (v 0 ) were obtained from the absorbance change of TMB according to the Lambert−Beer law (ε of 39 000 m cm −1 for oxTMB).…”
