Karyotype structures in representative taxa of the genus Allium L. are compared, based on Feulgen-stained somatic metaphase chromosomes. The karyotypes of Allium arlgirdense, A. anacoleum and A. microspathum (in subg. Allium), and A. rhetoreanum and A. shirnakense (in subg. Melanocrommyum) are described for the first time. The karyological analyses indicate the relationships among the species with respect to asymmetry indices. All Allium taxa studied are diploid with 2n = 2x = 16 chromosomes. One B chromosome is detected in the chromosomes of A. microspathum. All chromosome sets of the studied taxa consist of seven median chromosomes and one submedian chromosomes and all submetacentric pairs bear a satellite on the short arms. The lengths of the chromosomes show remarkable differences particularly at the subgenus level. The three subgenus of genus Allium show a clear tendency to have karyotypes distinct on asymmetry grounds. Karyotype asymmetry relationships are discussed according to the bidimensional scatter plots of A 1-A 2 and CV CL-M CA .

The onion plant (Allium cepa L.) is a suitable indicator plant for the determination of potential genotoxic agents in the samples taken from the environment. The genotoxic level of the agent under study is reflected by structural changes of the chromosomes and their changed numbers. The chromosomes under study are taken from the meristem cells of the young growing onion roots. A healthy normal onion cell has 16 (2n = 16) chromosomes. They are relatively large and so very appropriate for the detection of morphological changes. Prior to the chromosome study the root tip cells were immersed in a 0.1% aquatic solution of colchicine which stopped the mitotic cycle continuing beyond metaphase. The changes in morphology varied from a single distortion of a single chromosome up to several morphological changes observed on many chromosomes. We identified 15 categories of morphological aberrations which are classified into three groups: chromatid damage (CtD), centromere damage (CmD) and chromosome damage (CsD). CtD includes: single break chromatid, double break chromatid, isochromatid break, multiple break chromatid, gap chromatid, centric ring chromatid, acentric ring chromatid and triradial chromosomes. CmD includes: break centromere, gap centromere, single break centromere, double break centromere and multiple break centromere. CsD includes: ring chromosomes and dicentric chromosomes. Sometimes also the chromosome number changed which occurred as aneuploidy with monosomy 2n = 15 (2n =16-1) and euploidyincreased number of the basic chromosome number (2n = 6x-8x). We identified also the translocation: t(3p-; 5p+).

Allium is the largest genus of Amaryllidaceae comprising more than 900 species belonging to 15 subgenera. Iran is reported to be a center of diversity for subgenus Melanocrommyum. Acanthoprason is a section in this subgenus. In this paper, karyomorphological variations of nine populations of four species including Allium derderianum, A. kurdistanicum, A. minutiflorum and A. subakaka of the section Acanthoprason growing in Iran were undertaken using squash technique and 2% (w/v) aceto-orcein stain. All of the populations have the same chromosome numbers 2n = 2x = 16 with the exception in Kochka population of A. derderianum 2n = 18. In the present study, the chromosome number of A. minutiflorum and new chromosome number of A. derderianum were revealed for the first time. Chromosomal characteristics were determined using photographs complemented by cluster analysis. According to dendrogram generated by the unweighted pair-group method with arithmetic average (UPGMA) cluster analysis, nine populations studied were separated into five groups at a cut off value of 5. Karyotype analysis indicated that Allium species studied here generally have metacentric and submetacentric chromosomes and symmetric karyotypes. Results of the present study revealed the natural variation in nine populations of four species of Acantoprason section which can further serve conservation and breeding planning.
