We evaluated the potential of metabarcoding in assessing the environmental DNA (eDNA) biodiversity profile in the water column of an hydroelectric power plant reservoir in southeast Brazil. Samples were obtained in three technical replicates at 1 km from the dam at 1, 13 and 25 m depths. For each minibarcodes -- COI, 12S and 16S -- 1.5 million paired-reads (150 base pairs) were sequenced. A total of 44 unique taxa were found. COI identified most of the taxa (34 taxa; 77.2 %) followed by 16S (14; 31.8 %) and 12S (10; 22.7 %). All minibarcodes identified fishes (13 taxa), however, COI detected other aquatic macro-invertebrates (18), algae (3) and amoebas (2). Richness was the same across the three depths (35 taxa), although, beta diversity suggested slightly divergent profiles. In just one location we identified 15 taxa never reported previously, 50% of the fish species identified in the last year of fishery monitoring and 13% of the species in biodiversity surveys performed from 2012 to 2021. Clustering into Amplicon Sequence Variants (ASV) showed that 12S and 16S are able to detect predominant haplotypes of fishes, suggesting they are suitable to study population genetics of this group. In this study we reviewed the species occurring within the Três Irmãos reservoir according to previous conventional surveys and demonstrated that eDNA metabarcoding can be applied to monitor its biodiversity.

Esta pesquisa teve como objetivo verificar a quantidade e tipos de sedimentos transportados no sistema fluvial, a montante da Usina Hidrelétrica-UHE de Colíder, no médio curso do rio Teles Pires, região norte do Estado de Mato Grosso. Foram efetivados cinco campos no período de dois anos. Monitorou 4 (quatro) pontos para coleta de sedimentos de fundo e em suspensão. No trabalho em gabinete foram realizadas as análises do tamanho e forma dos grãos 2,00 mm. Na pesquisa em laboratório realizou-se a análise das amostras coletadas em campo pelo método de peneiramento e pipetagem. Por meio deste estudo verificou-se que o rio Teles Pires apresenta afloramento rochoso no leito e corredeiras. Fator este, que explica o aparecimento de sedimentos grossos no fundo do canal. Com a construção, instalação e funcionamento da usina pode-se averiguar que a carga sedimentar variou. Em algumas campanhas registrou maior concentração da composição arenosa, enquanto no enchimento do reservatório o silte teve valores elevados no fundo do canal, o que pode associar a submersão das margens, colmatagem dos afluentes e a perda da velocidade. A construção e funcionamento da UHE Colider contribuíram em mudanças significativas na fisiografia do canal e da paisagem no entorno.

In reservoir projects it is important to estimate when the accumulated sediments will start to interfere with their functions. However, predicting silting is difficult because the processes involved have some uncertainties. Thus, the study is not only deterministic, as currently performed, but also stochastic. Thus, the objective of this paper was to develop a stochastic method and evaluate its performance in estimating silting in reservoirs. The method has as originalities the fact of having coupled a deterministic model widely used in the area of Hydraulics to a stochastic one. Another originality was to validate the stochastic method developed from silting data obtained in the reduced model of a Small Hydroelectric Power Plant (SHP). Thus, it was observed that the real silting was always between the 1st and 3rd quartile of probability of the stochastic result. Thus, the main advantage of the stochastic model developed was to allow obtaining the probabilities of silted heights in the stretches of interest. In addition, the variability of the results in the simulations indicated the sections that may suffer greater silting. In this way, hydraulic structures can be better positioned. Preventive and corrective measures can also be better planned and executed.
