A Human Long Non-Coding RNA ALT1 Controls the Cell Cycle of Vascular Endothelial Cells Via ACE2 and Cyclin D1 Pathway

Abstract: Background/Aims: ALT1 is a novel long non-coding RNA derived from the alternatively spliced transcript of the deleted in lymphocytic leukemia 2 (DLEU2). To date, ALT1 biological roles in human vascular endothelial cells have not been reported. Methods: ALT1 was knocked down by siRNAs. Cell proliferation was analyzed by cck-8. The existence and sequence of human ALT1 were identified by 3’ rapid amplification of cDNA ends. The interaction between lncRNA and proteins was analyzed by RNA-Protein pull down assay, R… Show more

“…Non-coding RNAs are important cell cycle modulators and levels of miR-138, miR-195, and miR-206 have been predicted to promote CCND1 expression, which regulates the progression from G1 phase to S phase of the cell cycle (26) (27). There is evidence that long non-coding RNA ALT1 controls the cell cycle progression via ACE2 and Cyclin D1 pathway (28). In the present study, we report that the lncRNA CRYBG3 regulates cell division and its expression is dynamically associated with cell cycle progression, being lowest in M phase and highest in G1 phase.…”

“…Li and colleagues showed that lncRNA ALT1 directly interacts with ACE2 in HUVECs cells. ALT1 knockdown decreases ACE2 level thereby inhibiting cell proliferation via promotion of cyclin D1 ubiquitination 92 . Several studies have also addressed that lncRNAs control cell proliferation, autophagy, or colony formation through DPP4 93‐95 .…”

“…injections, Ang (1‐7) and A779 were dissolved in Ringer 's solution (Fuso Pharmaceutical Industries, Japan). The specificity of the anti‐Mas receptor and anti‐ACE2 antibodies used in this study has been demonstrated using Mas receptor knockout mice (Freund et al, ) and ACE2‐siRNA transfected cells (Li et al, ), respectively. The antibody against the membrane‐bound anti‐Na + /K + ‐ATPase used in our study has been reported to selectively bind the protein in membrane fractions but not in cytoplasmic fractions (Aravena et al, ; Planès et al, ).…”
