A highly sensitive and direct competitive enzyme-linked immunosorbent assay for the detection of di-(2-ethylhexyl) phthalate (DEHP) in infant supplies

Abstract: A sensitive and specific direct competitive enzyme-linked immunosorbent assay (dc-ELISA) was studied in this paper for the detection of di-(2-ethylhexyl) phthalate (DEHP) based on an antigen-coating format. The DEHP-specific polyclonal antibody was raised in rabbits and used to construct the dc-ELISA for the measurement of DEHP. The conjugates of the antibody with horseradish peroxidase (HRP) were used as the detection probe. Under optimal conditions, the assay had a detection limit (LOD) of about 0.0042 ng mL… Show more

“…We then validated the specificity between the anti-20 AgNs and anti-50 AgNP antibodies. The cross-reactivity determined using ELISA represented the specificity of antibodies, as described in a previous study by our groups . Surprisingly, although similar IgG titers were observed for the anti-20 and anti-50 nm AgNP antibodies, significant differences were recognized.…”

“…Additionally, the test time is 30 min, which is faster than current reported tests. 24,28 Overall, the results suggest that the designed aptasensor could be used for the sensitive detection of DEHP in standard sample.…”

“…For instance, ELISA assays were developed for sensitive detection of DEHP. [24][25][26] However, these immunoassays require costly antibodies and a long reaction time. Other sensors are proposed for detection of DEHP using functionalized glassy carbon electrodes 27,28 or extended gate organic eld effect transistor (EG-OFET), 29 however, these sensors show a low sensitivity in detection.…”

“…The Ren group has established a direct competitive immunoassay for the detection of mono-2-ethylhexyl phthalate in human urine (Feng et al, 2013). In addition, our group has reported several types of enzyme-linked immunoassay for detection of PAEs including di-(2-ethylhexyl) phthalate in infant supplies, dicyclohexyl phthalate in foods, dipropyl phthalate in plastic food contact materials, dimethyl phthalate (DMP) in water samples and diethyl phthalate (DEP) in spiking water samples Zhang et al, 2015;Zhang et al, 2011). These strategies enhance the efficiency and specificity of enzyme-based immunoassay.…”
