A Direct Redox Regulation of Protein Kinase C Isoenzymes Mediates Oxidant-induced Neuritogenesis in PC12 Cells

Abstract: In this study, we have used the PC12 cell model to elucidate the mechanisms by which sublethal doses of oxidants induce neuritogenesis. The xanthine/xanthine oxidase (X/XO) system was used for the steady state generation of superoxide, and CoCl 2 was used as a representative transition metal redox catalyst. Upon treatment of purified protein kinase C (PKC) with these oxidants, there was an increase in its cofactor-independent activation. Redox-active cobalt competed with the redoxinert zinc present in the zinc… Show more

“…Mitochondrial association of cell survival isoenzyme PKC⑀ likely protects cells from cell death induced by OGD/R. Our previous studies have shown an activation of ERK by ROS in GTPP-treated cells as well as an activation of ERK by PKC in oxidant-treated cells triggering downstream nuclear events (20,56). Based on these observations, we propose that GTPP induces expression of antioxidant enzymes that may precondition cells against OGD/R-induced cell death.…”

“…Stable Transfection of PKC⑀-We used previously generated PC12 cells stably transfected with either a metallothioneindriven PKC⑀ expression vector (to overexpress PKC⑀) or an empty vector (as a control) (56). Western blot analysis was used to determine the extent of expression of PKC⑀ in these transfectants.…”

“…In fact, a direct modulation of gCl by ROS can occur by involving oxidation of critical cysteine residues of ClC-1 channel, as described on heterologously expressed systems (2,79). PKC isoforms are also redox sensitive, and oxidation of accessory subunits has been shown to lead to a cofactor independent activation of PKC in PC12 cells (33). Therefore, a possible reinforcing loop generated by ROS on PKC activation is feasible and may also account for the ability of both NAC and chelerythrine to antagonize the ANG II-related increase of gK.…”

“…As early as 1979, May and de Haen demonstrated that H 2 O 2 acts as a "second messenger" during insulin stimulation (31), and in another study it was found that H 2 O 2 subsequently induces oxidative inactivation of the tyrosine phosphatase PTP1B to promote signal transduction (32). In addition, ROI have also been implicated in the regulation of transcription factors, such as AP-1 (1) and NF-〉 (30,38), and kinases such as protein kinase C (15). To prevent oxidative stress, cells must possess the means to regulate ROI in order to modulate signal transduction.…”
