A cytometric exercise in plant DNA histograms, with 2C values for 70 species

Abstract: An introduction is given to the literature concerning methods and objectives for cytometric DNA analysis of plant nuclei. This area has gained relevance with applications in plant breeding and seed production industries, where laboratories unfamiliar with cytometry are adopting the method. An extensive graphical guide to interpreting DNA histograms and their problems is given. Conversely, cytometry laboratories unfamiliar with plant sciences will find herein a guide, and references, to adapt their methods to p… Show more

“…The AT frequency was calculated from the DAPI factor with Eqs. (6) and (7). For the primary standard Pisum sativum, a value of 2C ϭ 9.07 pg and an AT frequency of 61.5% were used (see "Genome Size and AT Frequency of the Standards").…”

“…Doubled-haploid Pahang (DH-Pahang, ITC1511) was obtained from wild M. acuminata subspecies malaccensis accession 'Pahang' through anther culture and spontaneous chromosome doubling 31 . Genome sizes were estimated by flow cytometry according to Marie and Brown 32 . High molecular weight DNA was prepared from the youngest fully expanded leaf of DH-Pahang as described in Piffanelli et al 33 with minor modifications (Supplementary Text).…”

“…Estimation of genome size and ploidy. We estimated genome size by flow cytometry 55 and obtained an estimate of 800 Mb (Supplementary Table 2). We additionally estimated genome size by k-mer distribution analysis with the program Jellyfish 56 (k = 31), using Illumina short reads, and obtained an estimate of 738 Mb.…”
