A custom-made two-photon microscope and deconvolution system

Abstract: We describe in detail a custom-built two-photon microscope based on a modified confocal scanhead (Olympus Fluoview) and mode-locked Ti:sapphire laser (Coherent Mira 900). This system has internal detectors as well as external whole-field detection and an electrooptical modulator for blanking the beam on flyback and effecting fast changes in excitation intensity. This microscope can be used in deep, scattering samples for quantitative measurements with a wide range of fluorophores (GFP, fura, calcium green, cal… Show more

“…A custom-made two-photon laser scanning microscope (Majewska et al, 2000b) was used for in vivo imaging. The microscope consists of a modified Fluoview confocal scan head (Olympus Optical) and a titanium/sulphur laser providing 100 fs pulses at 80 MHz at a wavelength of 920 nm (Tsunami; Spectra-Physics, Menlo Park, CA) pumped by a 10 W solid-state source (Millenia; Spectra-Physics).…”

“…It consists of a modified upright Olympus microscope and a modified confocal scan head fitted with a ϫ60 water immersion objective (NA ϭ 0.9) and a Spectra-Physics Tsunami femtosecond laser tuned to 780 nm (26). A nondescanned photomultiplier tube mounted off the epifluorescence filter turret was used to detect fluorescence emission of Ͻ700 nm (12).…”

“…Two-photon microscopes suitable for imaging dendritic spines can be custom-built (e.g., Mainen et al 1999;Majewska et al 2000) or obtained from a commercial vendor (e.g., Olympus, Prairie Technologies, Sutter Instruments, Zeiss). A typical custom imaging setup for two-photon imaging of dendritic spines is shown in Figure 1.…”
