There is no project grant funding associated with this review.Non-alcoholic fatty liver disease (NAFLD) encompasses a spectrum of liver diseases, of which the first stage is steatosis. It is one of the most common liver diseases in developed countries and there is a clear association between type 2 diabetes (T2DM) and NAFLD. It is estimated that 70% of people with T2DM have NAFLD and yet there is currently no licensed pharmacological agent to treat it. Whilst lifestyle modification may ameliorate liver fat, it is often difficult to achieve or sustain; thus, there is great interest in pharmacological treatments for NAFLD. Metformin is the first-line medication in the management of T2DM and evidence from animal and human studies has suggested that it may be useful in reducing liver fat via inhibition of lipogenesis and increased fatty acid oxidation. Findings from the majority of studies undertaken in rodent models clearly suggest that metformin may be a powerful therapeutic agent specifically to reduce liver fat accumulation; data from human studies are less convincing. In the present review we discuss the evidence for the specific effects of metformin treatment on liver fat accumulation in animal and human studies, as well as the underlying proposed mechanisms, to try and understand and reconcile the difference in findings between rodent and human work in this area. The present study was performed to examine the effect of an artichoke and milk thistle consumption on liver function, kidney function, and lipid profiles. The chemical constituents were determined for the tested dried artichoke and milk thistle. This search was carried out using 72 adult male white albino rats randomly classified into two main groups. The first main group (36 rats) fed on a basal diet containing 20%-40% artichoke and 20%-40% milk thistle (acute liver failure groups). The second main group (36 rats) fed on a basal diet containing 20%-40% artichoke and 20%-40% milk thistle (acute renal failure groups).Then, the main groups were divided into six subgroups for each (n = 6). Results revealed that all groups treated with CCl4 or glycerol/ saline solution and administrated with different levels of dried artichoke and milk thistle (20%-40%) had a significant decrease in their liver function, kidney function, total cholesterol, triglycerides, LDL-c and VLDL-c cholesterol compared with the positive control group. On the other hand, a significant increase in HDL-c was recorded in all groups administrated with dried artichoke and milk thistle (20%-40%) compared with the control positive group. The results indicated that the consumption of artichokes and milk thistle could help in reducing the symptoms of acute liver and renal failure.

The present study is the first attempt to evaluate the hepatoprotective effect of ethanolic seeds extract (ESEt) of Centratherum anthelminticum (black cumin) in carbon tetrachloride (CCl 4)-induced liver injury. The test doses (600 and 800 mg/kg) of same extract were found effective in their respective test groups by improving the body and liver weights, serum alanine and aspartate transaminases, γglutamyltranspeptidase, alkaline phosphatase, total proteins, albumin, total bilirubin, especially indirect bilirubin and uric acid levels as compared to CCl 4-induced hepatotoxic control group. In addition, decreased percent inhibitions of antioxidant parameters including catalase, superoxide dismutase and reduced glutathione accompanied with increased percent inhibition of lipid peroxidation observed in both test groups. Histopathological studies also proved the liver regenerating property of ESEt by showing decrease in fatty deposition, necrosis and inflammation around the central vein of liver lobules. Therefore, the ESEt was found to be hepatoprotective and antioxidative in nature.
