A critical role for 14-3-3ζ protein in regulating the VWF binding function of platelet glycoprotein Ib-IX and its therapeutic implications

Abstract: The platelet receptor for von Willebrand factor (VWF), glycoprotein (GP) Ib-IX, mediates platelet adhesion and activation. The cytoplasmic domains of the GPIb ␣ and ␤ subunits contain binding sites for the phosphorylation-dependent signaling molecule, 14-3-3. Here we show that a novel membrane-permeable inhibitor of 14-3-3-GPIb␣ interaction, MP␣C, potently inhibited VWF binding to platelets and VWF-mediated platelet adhesion under flow conditions. MP␣C also inhibited VWF-dependent platelet agglutination induce… Show more

“…The cytoplasmic tail of GpIba interacts with proteins such as 14-3-3z and filamin, thus influencing cell signaling and actin cytoskeleton function, respectively (Asazuma et al, 1997;Kanaji et al, 2004;Dai et al, 2005;Ozaki et al, 2005). However, the transcriptional consequences of this are unclear.…”

“…MPalphaC, an inhibitor of 14-3-3zeta GPIbalpha interactions, which mediates von Willebrand factor binding to GPIb-IX, may be useful in treating or preventing thrombosis. 95 A nitric oxide-releasing aspirin, NCX-4016 Nitroaspirin, exerts a wide range of antiplatelet activity, superior antithrombotic activity, and reduces restenosis after arterial injury in animals. It has been more effective as an antiplatelet regimen with clopidogrel, as compared to aspirin.…”
