Due to their haploid nature, uniparental inheritance and apparent absence of recombination, organellar DNA markers are ideal tools for studying both intraspecific and interspecific phylogenies. Although 'universal' cytoplasmic DNA primers have become available for both red and green seaweeds, they are not transferable to brown seaweeds. Thus, based on the sequence of mitochondrial genome of the kelp Laminaria digitata, we developed universal-type primers for seven intergenic spacer loci and one mononucleotide tandem repeat locus. We investigated the conservation and sequence variation of these loci among nine Laminarialean and three fucoid species, as well as intraspecific polymorphism within five of these species. Six loci were conserved in at least four of the five brown algal families tested and five were polymorphic within at least one species. Although overall levels of divergence were relatively homogenous ( ¼ 0.314 AE 0.031), a hierarchical analysis suggests there is variation in the rate of evolutionary change among the six intergenic spacers. Due to their high degree of transferability, versatility and to non-coding nature, we argue that these intergenic spacer loci are highly efficient markers for within-family phylogenetic studies and for intra-specific phylogeographic studies in brown seaweeds.

We present an annotated checklist of the benthic marine and brackish algae of the Galician coasts (Spain) based on literature records and new collections. This checklist includes 618 species: 118 Cyanophyta, 296 Rhodophyta, 127 Ochrophyta, and 77 Chlorophyta. The number of specific, infraspecific taxa, and stages is 643: 121 Cyanophyta, 309 Rhodophyta, 135 Ochrophyta, and 79 Chlorophyta. Hyella caespitosa var. nitida, Calothrix fasciculata, Gracilariopsis longissima, Compsonema minutum, and Sphacelaria tribuloides are new records for Galicia, and there are also some new provincial records. We state the presence of each species for Lugo (Lu), A Coruña (Co), and Pontevedra (Po) provinces. The number of species found in Galicia is high, since 85% of the species recorded for the warm-temperate NE Atlantic Ocean grow in Galicia. Biogeographical comments comparing the Galician data with the neighboring areas of Britain and Ireland, Basque coast, Portugal, southern Iberian Peninsula, Canary Islands and Atlantic coast of Morocco are given. Finally, we present lists of cold-temperate, warm-temperate, Lusitanic Province endemics, and alien species growing in Galicia.

In brown algae (Phaeophyceae), only a few taxa are reported to have plastids containing pyrenoids and this character has often been used for systematic delineation. Members of the Ectocarpales display large, stalked and exserted pyrenoids in their plastids, whereas the Scytothamnales, as well as the genera Asterocladon, Asteronema and Bachelotia, exhibit plastids arranged in a stellate configuration with pyrenoids embedded in the plastid stroma. The evolution of plastid characters has remained difficult to establish, especially because of a persistent lack of resolution in brown algal molecular phylogenies. Here, we generated a seven-locus dataset for a species-rich taxon sampling, to reassess phylogenetic relationships of pyrenoid-bearing brown algal taxa. Within the Ectocarpales, the relationships among families are clarified. The minute chordariacean genus Herponema is assigned to the family Acinetosporaceae. The systematic positions of two enigmatic genera are revised: Chordariopsis is assigned to the Adenocystaceae, and Spongonema is transferred back to the Ectocarpaceae. The new order Asterocladales, which is sister to the Ectocarpales, is proposed to accommodate species of the genus Asterocladon. The order Scytothamnales is shown to include the genus Bachelotia in a new family Bachelotiaceae, as well as another new family Asteronemataceae, accommodating the genus Asteronema. These new taxonomic insights also shed light on the evolution of plastid morphology and arrangement in brown algae. Stellate plastid configurations have evolved independently in Asterocladales and Scytothamnales. Evolutionary scenarios underlying the diversity of stellate configurations in the Scytothamnales and the Asterocladales-Ectocarpales lineage are discussed.
