Human milk (HM) is extremely important to the baby, containing a varied and balanced range of nutrients fundamental to the ideal development of the newborn. Among these nutrients, HM lipids provide 40-60% of the energy. In HM banks, the creamatocrit method is employed as a quick and solvent-free tool, however, since Folch methodology is considered reference for lipid extraction, this study compared the fatty acid (FA) composition and the lipid profile by gas chromatography with flame ionization detector (GC-FID) and direct infusion by electrospray ionization mass spectrometry (DI-ESI-MS) in pools of different HM phases, obtained from Folch and creamatocrit methods, in order to elucidate which method provides better information about HM lipids. Lipid compositon results by Folch were superior to the results obtained by creamatocrit in HM pools investigated. It means, the lipid profiles of all HM pools were more intense, and the number of FA identified by Folch was also higher in comparison to the creamatocrit method (21 and 6 FAs, respectively). GC-FID analysis in combination with principal component analysis (PCA) divided the methods, confirming a greater contribution of the Folch method. Therefore, Folch methodology revealed an effective lipid extraction in comparison to creamatocrit method.

Despite the global increasing demand for goat milk, no data exist on the nutrient potential of Laoshan goat milk and limited information exists on its composition according to lactation period. In this study, the measured indicators, including dry matter, protein, immunoglobulin, lactoferrin, fat, ash, lactose, calcium, titratable acidity and pH from partum to 261 days postpartum of Laoshan goat milk, followed the same law of change. There was a significant difference over the first 5 days postpartum, after which no significant difference was observed until days 225 postpartum. However, days 255 postpartum was like a turning point, after which their values showed either an increasing or decreasing trend. We concluded that the colostrum for Laoshan dairy goat was considered from days 1 to 5 postpartum and after 5 days it was as transitioned to milk when all measured parameters were according to the normal ranges as described in this study. This work provided updated information on the current nutritive characteristics of Laoshan goat milk, which may be of importance in the fields of milk production, food technology, and nutritional mutual replenishment.

The supercritical fluid chromatography coupled with mass spectrometry (SFC-MS) method and liquid chromatography coupled with mass spectrometry (LC-MS) method were developed for the separation and characterization of poly (ethylene oxide) methyl glucose sesquistearate (PEO-Glu-sesquistearate). The products of PEO-Glusesquistearate are composed of complex oligomers. The relationship between molecular structure of these oligomers and chromatographic retention behavior in both SFC and LC were discussed and compared. As compared with LC, hydrophobic moieties of compounds favor the fast elution in SFC. The different series can be better separated by LC, while the homologues compounds in same series can be better separated by SFC, and SFC-MS provided more comprehensive structural information. Different series such as PEO-distearate, PEO-stearate, PEO, PEO-Glu-tetrastearate, PEO-Glu-tristearate, PEO-Glu-distearate, PEO-Glu-stearate, and PEO-Glu were identified by MS/MS.
