A capillary electrophoresis coupled to mass spectrometry pipeline for long term comparable assessment of the urinary metabolome

Abstract: Although capillary electrophoresis coupled to mass spectrometry (CE-MS) has potential application in the field of metabolite profiling, very few studies actually used CE-MS to identify clinically useful body fluid metabolites. Here we present an optimized CE-MS setup and analysis pipeline to reproducibly explore the metabolite content of urine. We show that the use of a beveled tip capillary improves the sensitivity of detection over a flat tip. We also present a novel normalization procedure based on the use … Show more

“…used a beveled tip sheath‐liquid interface, which was first developed by Tseng et al. , instead of a conventional sheath‐liquid interface . When applied to the analysis of pooled human urine samples, this CE‐MS approach provided the detection of slightly more metabolite features as compared to the standard CE‐MS method (Fig.…”

“…One of the long-standing issues with CE-MS is the ease and stability of the CE to MS interface. Ramautar [123] and Boizard [120] published on their work to improve the reliability and sensitivity of the MS interface. Ramautar provided data on using a sheathless capillary interface to improve the sensitivity with profiling glioblastoma cell lines, while Boizard designed a beveled capillary tip to improve the stability of the CE-MS interface, examining one sample over 130 times in 4 years.…”
