Aedes aegypti mosquitoes, main vectors for numerous flaviviruses, have olfactory preferences and are capable of olfactory learning especially when seeking their required environmental conditions to lay their eggs. In this study, we showed that semiochemical conditions during Aedes aegypti larval rearing affected future female choice for oviposition: water-reared mosquitoes preferred to lay eggs in water or p-cresol containers, while skatole reared mosquitoes preferred skatole sites. Using two independent behavioural assays, we showed that this skatole preference was lost in mosquitoes infected with dengue virus. Viral RNA was extracted from infected female mosquito heads, and an increase of virus load was detected from 3 to 10 days post infection, indicating replication in the insect head and possibly in the central nervous system. Expression of selected genes, potentially implied in olfactory learning processes, were also altered during dengue infection. Based on these results, we hypothesise that dengue virus infection alters gene expression in the mosquito’s head and is associated with a loss of olfactory preferences, possibly modifying oviposition site choice of female mosquitoes.

Insects perceive the environment with a repertoire of promiscuous and narrowly tuned odorant receptors (ORs). The southern house mosquito, Culex quinquefasciatus, utilizes two ORs, CquiOR10 and CquiOR2, which are narrowly tuned and well conserved among mosquito species. They detect the oviposition attractants skatole and indole, respectively, with reciprocal specificity. We envisioned that by reverting specificity with chimeric receptors, we could gain a better insight into the specificity determinants of insect ORs. We swapped the seven transmembrane (TM) domains of CquiOR10 and CquiOR2 and identified TM2 as a specificity determinant. With additional mutations, we showed that CquiOR10A73L behaved like CquiOR2. Conversely, CquiOR2L74A recapitulated CquiOR10 specificity. Next, we generated structural models of CquiOR10 and CquiOR10A73L using RoseTTAFold and AlphaFold and docked skatole and indole using RosettaLigand. These modeling studies suggested space-filling constraints around A73, although this residue in CquiOR10 did not directly form contacts with skatole or indole. Consistent with this hypothesis, CquiOR10 mutants with a bulkier residue (Ile, Val) were insensitive to skatole and indole, whereas CquiOR10A73G retained the specificity to skatole and showed a higher sensitivity than the wild-type receptor CquiOR10. On the other hand, Leu to Gly mutation of the indole receptor CquiOR2 reverted the specificity to skatole. Lastly, CquiOR10 was insensitive to 3-ethylindole, whereas CquiOR10A73G responded to the three ligands with 3-ethylindole eliciting a slightly higher response than indole. We then concluded that the specificity of these receptors is mediated by a single amino acid substitution leading to dilation or contraction of the binding pocket.

Despite the increasing advocacy towards the "openness" of science and research data, it is still far from being a widespread practice. The goal of this paper is to identify the most pressing obstacles (in terms of funding, technology, Intellectual Property Rights, contracts, data protection, and social norms), which are hindering the development of Open Science and Open Research Data, with particular attention to the situation of developing countries. The innovative aim of this paper, which is the first essay of a broader research, is to prepare the epistemological basis for a Law and Technology theory of "Open Bioinformation" (OB), where bioinformation stands for research data in life sciences. We argue that so far the literature has addressed the promotion of openness in science and research data only in a sectorial manner, taking into account just one or a few of the factors affecting openness as if they were not related or mutually influenced. Therefore, the suggested solutions are limited to a single perspective and fail to consider the dynamics of information control. In our view, a holistic approach, that tries to zoom out from the specific disciplines and take into account the whole picture, would contribute to determining an effective policy for promoting OB. For this reason, we have to consider the technological, legal, and sociological aspects, in order to assess whether and how changes in one domain might affect the others.
