8-Oxo-7,8-Dihydro-2′-Deoxyguanosine (8-oxodG) and 8-Hydroxy-2′-Deoxyguanosine (8-OHdG) as a Potential Biomarker for Gestational Diabetes Mellitus (GDM) Development

Abstract: The growing clinical and epidemiological significance of gestational diabetes mellitus results from its constantly increasing worldwide prevalence, obesity, and overall unhealthy lifestyle among women of childbearing age. Oxidative stress seems to be the most important predictor of gestational diabetes mellitus development. Disturbances in the cell caused by oxidative stress lead to different changes in biomolecules, including DNA. The nucleobase which is most susceptible to oxidative stress is guanine. Its da… Show more

“…The last one results from the binding of the ● OH to the eighth carbon of the guanine [ 96 ]. 8-OHdG transforms into its oxidised form 8-oxo-7,8-dihydro-2′-deoxyguanosine (8-oxodG) through a keto-enol tautomerism reaction, which is considered the most abundant oxidative injury, with about 100,000 8-oxodG generated every day in a single cell [ 97 , 98 ].…”

“…Promotion of oxidation of protein intensifies during GDM, in which the hyperglycemic state causes protein hydroperoxides, protein carbonyls, C-reactive protein and glycated hemoglobin (HbA1c). In addition to this, it is considered an important mediator of adipocyte disorders, intensifying the inflammatory response and contributing to the complications of diabetes 30 .…”

“…Recently it has been speculated that 15-F 2t -IsoP excretion is associated with 7,8-dihydro-8-oxo-2′-deoxyguanosine (8-oxo-dGuo or 8-oxo-dG) [ 19 , 20 ], a guanine modification induced by free radicals and adopted as a non-invasive biomarker of OS [ 21 ]. The accumulation of such damages can lead to genetic information changes, and, consequently, to mutagenesis and cells apoptosis [ 22 ]. The urinary concentration of this biomarker reflects the equilibrium between its production and repair in both DNA and the nucleotide pool [ 23 ].…”
