Current knowledge point to persistence of risk factors for the development of cervical intraepithelial neoplasia. The infection with a high-risk oncogenic Human Papillomavirus (HPV) subtypes, most commonly 16 and 18, is a necessary, although not sufficient, condition for development of invasive cervical cancer (ICC) and its precancerous precursor, cervical intra-epithelial neoplasia (CIN). It has been suggested that CIN disease severity and the diversity of vaginal microbiota are associated and this may determine viral persistence and disease behaviour. Our work focuses on the genetic variability associated to the modulation of genotoxicity induced by vaginal microbiota diversity. Relatively little is known about the mechanisms associated with clearance or persistence of HPV infection, therefore we hypothesized that may be under the influence of the genetic background.

Nitric oxide (NO) plays complicated roles in carcinogenesis. Endothelial nitric oxide synthase (eNOS) gene is responsible for most of the NO produced. For this reason, it was considered that the eNOS gene variants is associated with cancer suspectibility. The aim of this study was to determine whether eNOS variants (G894T and intron 4 VNTR a/b) affect in Chronic Lymphocytic Leukemia (CLL) risk in Turkish patients. This is a prospective single-center crosssectional study in patients with CLL. A total of 60 CLL patients and 100 healthy controls with similar age and sex were included to this study. Two eNOS gene variants (G894T and intron 4VNTR a/b) were analysed with polymerase chain reaction (PCR) and/or restriction fragment length polymorphism (RFLP) methods. In this study, we found that the TT genotype of eNOS G894T variant was significantly associated with an increased risk in patient with CLL compared with control (OR: 0.867, Cl: 0.785-0.957, p= 0.001). There was not any significant difference in the eNOS G894T allele distribution between the groups (p> 0.05). In addition, no significant difference was detected between the CLL patients and healthy controls with respect to the frequencies of genotypes and alleles in intron 4 VNTR a/b variant of eNOS. eNOS gene variants (G894T and intron 4 VNTR a/b) in CLL patients were simultaneously analyzed for the first time in present study. Our study suggest that the eNOS G894T variant may be associated with the development of CLL in the Turkish population.

Breast cancer is the leading cause of death in women. Prognosis of breast cancer is often pessimistic because the tumors are prone to metastasizing to the bone, brain, and lung. The estrogen signaling receptor (ESR) pathway contains 39 main genes and proteins which makes it one of the larger signaling pathways. Predominately this pathway and the proteins within are involved in breast growth and development, making it a prospective area of study for breast cancer. While the healthy ESR pathway has been constructed and is well established, a mechanistic model of mutated genes of ESR pathway has not been delved upon. Such mutated models could be utilized for selecting combinational targets for drug therapies, as well as elucidating crosstalk between other pathways and feedback mechanisms. To construct the mutated models of the ESR pathway it is imperative to assess what is currently understood in the literature and what inconsistencies exist in order to resolve them. Without this information, a model of the ESR pathway will be unreliable and likely unproductive. This review is the detailed literature survey of the biological studies performed on ESR pathways genes, and their respective roles in breast cancer. Furthermore, the details mentioned in the review can be beneficial for the integrated study of the ESR pathway genes, which includes, structural and dynamics study of the genes products, to have a holistic understanding of the cancer mechanism.
