3H-imipramine and3H-cyano-imipramine binding in rat brain tissue: Effect of long-term antidepressant administration

Abstract: 3H-imipramine and 3H-cyano-imipramine binding was determined in brain homogenates of rats which had been treated for 21 days with imipramine or desimipramine. When compared to control animals, long-term administration of these antidepressants did not induce any alteration in the maximal number of 3H-imipramine or 3H-cyano-imipramine binding sites. However, a transient increase in the apparent dissociation constant was observed. Such findings are discussed in respect to previous studies, which have been highly … Show more

“…Controversial published data have shown that chronic treatment with different antidepressants can produce either a down-regulation Racagni et al, 1983;Barbaccia et al, 1983) or no change (Plenge and Mellerup, 1982;Lee et al, 1983;Gentsch et al, 1984) in [3H]IMI recognition sites.…”

“…Other reports show that imipramine and desipramine produce down-regulation of desipramine displaceable [3H]imipramine binding sites in rat cortex (Raisman et al 1980) and hippocampus (Barbaccia et al 1983). But other studies report on no alterations in cortical binding after chronic imipramine or desipramine treatment (Plenge and Mellerup 1982;Gentsch et al 1984;Abel et al 1985;Biegon 1986). However, reduced binding (and higher Ka values) due to inhibition by remaining uptake inhibitor in the tissue preparation when no or short withdrawal intervals were used, has been observed (Plenge and Mellerup 1982;Wilson and Roy 1985).…”
