The roles of endothelial nitric oxide synthase gene polymorphisms in diabetes mellitus and its associated vascular complications: a systematic review and meta-analysis

Abstract: In conclusion, our findings indicate that rs891512, rs1799983, rs2070744, and rs869109213 polymorphisms may serve as genetic biomarkers of DM, while rs1799983, rs2070744, and rs869109213 polymorphisms may contribute to the development of vascular complications in DM.

“…Third, associations between ANRIL variants and IS may also be modified by gene–environment interactions. However, most eligible studies ignore these potential interactions, which impeded us to perform relevant analyses accordingly [35,36]. On account of above-mentioned limitations, our findings should be cautiously interpreted.…”

“…In a Chinese Han, the rs2070744 C allele was associated with an increased risk of CAD [22]. In patients with diabetes mellitus, this allele was also associated with the risk of CAD [28]. Alternately, a multivariate analysis by Kincl et al [29] did not confirm any significant associations between the rs2070744 polymorphism and solitary CAD or CAD in combination with obesity and diabetes mellitus.…”

“…Additional analysis of BMI, waist circumference, and HDL cholesterol concentrations, performed separately in subgroups of females and males, showed significantly higher BMI values and waist circumferences in females with the rs1799983 TT vs. GT + GG genotype (median value [interquartile range]: 32 [31][32][33][34][35] vs. 27 [25][26][27][28][29][30] kg/ m 2 , p = 0.007, and 103 [98-106] vs. 92 [84-100] cm, p = 0.018, respectively). There were no associations between the above parameters and rs1799983 polymorphism in males.…”

“…This additive interaction could be due to the combination of the destructive effects on renal circulation exerted by smoking [17] and impaired renal haemodynamic balance, which is associated with KCNQ1 rs2237895 [22]. Obesity is associated with endothelial injury [23] and, coupled with vasoconstriction from reduced eNOS activity due to eNOS rs 2070744 [24], could cause a double hit on renal function.…”
