3-Deazaadenosine mitigates arterial remodeling and hypertension in hyperhomocysteinemic mice

Abstract: Chronic hyperhomocysteinemia (HHcy) is an important factor in development of arterial hypertension. HHcy is associated with activation of matrix metalloproteinases (MMPs); however, it is unclear whether HHcy-dependent extracellular matrix (ECM) accumulation plays a role in arterial hypertrophy and hypertension. We tested the hypothesis that in HHcy the mechanism of arterial hypertension involves arterial dysfunction in response to ECM accumulation between endothelial and arterial smooth muscle cells and subseq… Show more

“…Experiments using CBS knockout mice were not considered because they have serious hyperhomocysteinemia (40-to 50-fold elevated plasma homocysteine) and need to be treated with homocysteine-lowering drugs to survive a reduced life span (33)(34)(35)(36). These mice have growth retardation, reduced survival and altered vascular responsiveness to cholinergic and bradykinin stimulation, making their use of questionable value in the present experiments.…”

“…Several studies have demonstrated vascular hypertrophy, remodeling, altered vascular mechanics, or increased stiffness of arteries or arterioles in mice with hyperhomocysteinemia. 39,49,82,83 Increased neointima formation in response to arterial injury also has been observed. 38 Accelerated thrombosis of the carotid artery has been detected using the rose bengal photochemical thrombosis method 33 in Cbs…”

“…The present study has also shown the positive correlation between serum homocysteine and total cholesterol, Triglyceride, LDL cholesterol & negative correlation between serum homocysteine and HDL cholesterol in hypertensive subjects. Hyperhomocysteinemia, through its oxidative effects could reduce vasodilators like nitric oxide as well as promote extracellular matrix accumulation and smooth muscle cell proliferation, leading to vascular constriction and stiffness 36,37 . The atherogenicity of homocysteine may involve several mechanisms including LDLcholesterol oxidative modification, and HDL-cholesterol decrease 38 .…”
