The Ashitaba (Angelica keiskei) Chalcones 4-hydroxyderricin and Xanthoangelol Suppress Melanomagenesis By Targeting BRAF and PI3K

Abstract: Malignant melanoma is an aggressive tumor of the skin and still lacks effective preventive and therapeutic treatments. In melanoma, both the BRAF/MEK/ERK and PI3-K/AKT signaling pathways are constitutively activated through multiple mechanisms, which result in cell-cycle progression and prevention of apoptosis. Therefore, the development of novel strategies for targeting BRAF and PI3K are of utmost importance. In this study, we found that Ashitaba () chalcones, 4-hydroxyderricin (4HD) and xanthoangelol (XAG), … Show more

“…Natural 4-hydroxyderricin and xanthoangelol, in addition to inducing apoptosis in human melanoma cells, suppressed their growth by targeting BRAF and PI3K pathways. Moreover, treatment of melanoma cells with both chalcones led to a significant inhibition of phosphorylated Akt, downstream of PI3K in a dose-dependent manner [ 119 ].…”

“…Specifically, in the research field of anti-cancer effects, it is reported to show anti-osteosarcoma effect by inhibiting the activation and differentiation of M2 macrophages [ 37 ]. Moreover, another study demonstrates that 4-HD can suppress melanomatogenesis by targeting BRAF and PI3K [ 38 ]. Currently, the literature relating the inhibitory effect and mechanisms of 4-HD on liver cancer is limited.…”
