Layer‐by‐layer buildup of polysaccharide‐containing films: Physico‐chemical properties and mesenchymal stem cells adhesion

Abstract: Layer-by-Layer assembled polyelectrolyte films offer the opportunity to control cell attachment and behavior on solid surfaces. In the present study, multilayer films based on negatively charged biopolymers (pectin, dextran sulfate, carboxymethylcellulose) and positively charged polysaccharide chitosan or synthetic polyelectrolyte polyethyleneimine has been prepared and evaluated. Physico-chemical properties of the formed multilayer films, including their growth, morphology, wettability, stability, and mechani… Show more

“…It could be assumed that such cell behavior can be dealt with mechanical properties of film surface or toxic effect of residual GA on MSCs. Many researchers have previously shown that one of the key parameter effects on MSCs adhesion is the stiffness of film surface 12,13,49‐51 . The elastic properties of film surface can be quantitatively evaluated by PeakForce QNM (force volume mode).…”

“…Isolation of MSCs was performed by enzymatic treatment of Wistar rat fat homogenate in accordance with the previously reported procedure 13 . Briefly, after treatment of fat in the 0.1% collagenase solution (30 min, 37°C) the cell suspension was filtered through a capron filter (100 μm) and centrifuged at 370 × g for 10 min.…”

“…Despite the numerous works report the effect of polysaccharide addition on mechanical and biological properties of gelatin films, however, information about the stability and cellular affinity in high humidity environment is still limited. The improvement of stability and cellular affinity is important to maintain the integrality of films acting as skin barriers and support the repair process of stem cells (Afewerki et al, 2018; Dash et al, 2018; Kulikouskaya et al, 2018). This study prepared two kinds of gelatin‐polysaccharide composite films, including gelatin‐HA (G‐HA) and gelatin‐CS (G‐CS), and investigated the differences in the stability and cellular affinity between these two composite films.…”

“…При клеточной трансплантации отмечены сходные с альгинатами свойства [19]. Кроме того, тонкие пленки на основе пектина пригодны для эффективной адгезии МСК [20]. Композиционные полимер-неорганические материалы на основе пектинов, например нанокомпозиты пектин-Ag, проявляют собственную антибактериальную активность и обеспечивают синергетический антибактериальный эффект в сочетании с аминогликозидными антибиотиками [20].…”
