Functional analysis of the global repressor Tup1 for maltose metabolism in Saccharomyces cerevisiae: different roles of the functional domains

Abstract: BackgroundTup1 is a general transcriptional repressor of diverse gene families coordinately controlled by glucose repression, mating type, and other mechanisms in Saccharomyces cerevisiae. Several functional domains of Tup1 have been identified, each of which has differing effects on transcriptional repression. In this study, we aim to investigate the role of Tup1 and its domains in maltose metabolism of industrial baker’s yeast. To this end, a battery of in-frame truncations in the TUP1 gene coding region wer… Show more

“…Cyc8 (homologous with Ssn6 or SsnF) and Tup1 (homologous with RcoA) are critical transcriptional repressors in CCR. They are involved in nutrient uptake, mating type, and other metabolisms in S. cerevisiae [ 14 ]. In yeast, the recruit proteins Cyc8 and Tup1 are recruited by Mig1, and regulate the target’s expressions by binding to the promoter regions of downstream genes, such as diverse hydrolase genes [ 15 ].…”

“…Notably, the similar profiles of DAPs shared between mutants cyc8 Ϫ/Ϫ and tup1 Ϫ/Ϫ , for instance, showing upregulated glycolysis and gluconeogenesis proteins and downregulated amino acid biosynthesis proteins, suggest opposite and collaborative roles according to the environmental conditions. Indeed, it has been reported previously that the Cyc8-Tup1 complex showed antagonistic and activator effects (26)(27)(28), but it is necessary to elucidate this behavior and its effect on carotenogenesis.…”

“…Tup1 plays a role in histone deacetylation and chromatin remodeling in other contexts [ 21 , 28 ]; however, it is unclear whether Tup1 contributes to these processes in quiescence. Tup1 has been identified as a gene that is essential for viability of cells in G0 and is implicated in glucose repression in yeast, but there is no molecular mechanism to explain its role in these processes [ 42 46 ]. Here, we demonstrate that Tup1 is essential for repressing target genes in stationary phase by interacting with Rpd3 and Isw2 to generate regions of repressed chromatin.…”
