In Vivo Functional Selection Identifies Cardiotrophin-1 as a Cardiac Engraftment Factor for Mesenchymal Stromal Cells

Abstract: These results support the feasibility of selecting molecules in vivo for their functional properties with adeno-associated viral vector libraries and identify cardiotrophin-1 as a powerful cytokine promoting cell engraftment and thus improving cell therapy of the infarcted myocardium.

“…Ctf1 is a member of the IL-6 superfamily, and several studies have demonstrated that administration of Ctf1 can effectively reduce myocardial ischemia and reperfusion injury (41). Recently, it was reported that Ctf1 promoted MSC adhesion depending on activation of focal adhesion kinase and signal transducers and activators of transcription 3 (17). Indeed, we found that Ctf1 pretreatment further improved the survival of MSCs and the outcomes of MSC transplantation with DSAP.…”

“…Ctf1 is an IL-6 family member originally discovered as a prohypertrophic cytokine. It was reported that Ctf1 promoted MSC adhesive properties depending on activation of focal adhesion kinase (17). We found that the number of adherent MSCs pretreated with 200 ng/ml rCtf1 on the DSAP was significantly higher than that of the controls (Fig.…”

“…CT-1 is another member of the IL-6 family of cytokines and the most potent growth factor involved in myocardial hypertrophy (28). In recent studies, a role of CT-1 in extracellular matrix organization has been suggested (29).…”

“…Paradoxically, pretreating MSCs with the IL-6 family member cardiotrophin-1, prior to intramyocardial injection into infarcted mice, improved engraftment and infarct size reduction. 105 This result suggests that the host immune system itself helps to maintain MSC immunoprotection. This interplay was further illustrated by ixmyelocel-T cells, a mixed population of bone marrow mononuclear cells enriched for CD90 + MSCs (50-fold) and CD45 + CD14 + M2 macrophages ($200-fold).…”
