Xanthophyll esters are found in human colostrum

Abstract: Xanthophyll esters are significant contributors to the carotenoid profile in the colostrum, while mature milk does not contain these compounds. Therefore, fatty acid acylation to xanthophylls is activated during the accumulation of carotenoids in the human mammary gland. The sharp decline in the amount of xanthophyll esters in mature milk indicates that the lipophilic components are those recently incorporated in the mammary epithelium.

“…Carotenoids are considered to be normally present only in their free forms in human plasma. Although the accumulation of carotenoid esters has previously been reported in human skin, and most recently in human colostral milk, small amounts of carotenoid esters that might have been briefly present in human plasma TRL fractions . It has been hypothesized that the cleavage step takes place in the intestinal phase of digestion.…”

“…Carotenoids are universally present components in the human diet and are known to be present in colostrum and mature milk from mothers . The U.S. Food and Drug Administration has repeatedly accepted the Generally Recognized As Safe declarations made by supplement manufacturers for several carotenoid‐rich products, including palm oil carotenoids, several lutein‐containing, zeaxanthin‐containing, and lycopene‐containing products, some of them tomato‐based and also contain the colorless carotenoids phytoene and phytofluene…”

“…Safety of Carotenoids: Carotenoids are universally present components in the human diet and are known to be present in colostrum and mature milk from mothers. [108][109][110][111] The U.S. Food and Drug Administration has repeatedly accepted the Generally Recognized As Safe declarations made by supplement manufacturers for several carotenoid-rich products, including palm oil carotenoids, [112] several lutein-containing, [113][114][115][116][117][118] zeaxanthin-containing, [119][120][121] and lycopene-containing products, some of them tomato-based and also contain the colorless carotenoids phytoene and phytofluene. [122][123][124][125] Notably, dietary carotenoids are safe at concentrations much higher than those that are usually achievable in humans conforming to normal dietary patterns.…”

“…Therefore, the extraction methods mean the application of similar protocols applied for the extraction of free carotenoids, but the oily-dense nature of some matrices containing carotenoid esters requires clean-up steps. Different approaches have been set up, including the utilization of open column chromatography on silica gel [17] or magnesium oxide [18], enzymatic hydrolysis [19], solid-phase extraction [20], or even the use of semi-preparative isolation on C18 column and subsequent identification [21].…”

“…Another significant improvement in the hardware configuration of the mass spectrometer is the incorporation of a second stage of m/z analysis to obtain the mass spectra of selected ions. Thus, the arrangement of mass spectrometers in a tandem configuration to structure hybrid mass spectrometers (Q-time-of-flight, Q-IT, and triple Q-Orbitrap) [2,20,64,[83][84][85][86][87] allows the achievement of higher performance by increasing the mass accuracy level and the record of mass spectrum of a precursor ion to generate product ions. This configuration is essential when soft ionization methods are applied because they barely produce fragment ions and the experimental information from the MS analysis would limit to m/z values and isotopic pattern.…”
