Anti-aging effect and gene expression profiling of dung beetle glycosaminoglycan in aged rats

Abstract: BackgroundThis study aimed to evaluate the anti-aging effect of a newly prepared insect-derived compound, dung beetle glycosaminoglycan (GAG), given intraperitoneally to old SD rats as part of their diet for 1 month. Insect GAG administration was found to be related to a reduction in oxidative damage, hepato-cellular biomarker levels, protein carbonyl content, and malondialdehyde concentration. The anti-aging-related molecular genetic mechanisms of dung beetle GAG are not yet fully elucidated. Results Catharsiu… Show more

“…These insect N-glycans were identified polymer with hexose and N-acetylhexose in this study. Monosaccharide compositions of CaG [8] and IQG [9] have been reported. N-glycan of CaG was identified as Hex 3 (Hex = arabinose or rhamnose) HexNAc 3 dHex 1 by MS/MS spectrometry.…”

“…Total RNA was isolated from melanoma tissue of melanoma mice treated with sample GAG. Complementary DNA (cDNA) was synthesized from 1 μg of total RNA to according to previously reported procedures [8]. For detection of target gene transcripts, we designed specific forward and reverse oligonucleotide primers using Beacon Designer software (PREMIER Biosoft, Palo Alto, CA, USA).…”

“…Therefore, we designed this study to determine whether insect glycosaminoglycans might have a role in treating diabetes or increasing antioxidant enzyme activity under diabetes condition using, Homo diabetic (db) mice. An insect glycosaminoglycan was been prepared and purified from black and scarlet cicada ( Huechys sanguinea ) ( 6 ). Another insect glycosaminoglycan isolated from dung beetle ( Catharsius molossus, Ca) also possesses anti-aging activities.…”

“…Protein carbonyl level and induction of antioxidant enzyme ratios following GAG treatment of db/db mice were also analyzed. Dung beetle (Catharsius molossus, Ca) GAG known to exhibit marked anti-ageing activity characterized by reduced oxidative damage in aged rats [16] was used as a positive control. In this work, we have also characterized purified N-glycans for deglycosylation of GbGAG followed by MS and MS/MS (MALDI TOF MS) analysis.…”
