Reduction by coffee consumption of prostate cancer risk: Evidence from the Moli‐sani cohort and cellular models

Abstract: Meta-analytic data on the effect of coffee in prostate cancer risk are controversial. Caffeine as a bioactive compound of coffee has not yet been studied in deep in vitro. Our study aimed at evaluating in a population cohort the effect of Italian-style coffee consumption on prostate cancer risk and at investigating in vitro the potential antiproliferative and antimetastatic activity of caffeine on prostate cancer cell lines. 6,989 men of the Moli-sani cohort aged ≥50 years were followed for a mean of 4.24 ± 1.… Show more

“…Notably, adjusting for social class and age, higher coffee consumption (three or more cups of coffee) was associated with significantly reduced risk of high Gleason grade prostate cancer compared with noncoffee drinkers [70]. Similarly, in a population cohort study, men with highest coffee consumption (>3 cups per day) had a 53% lower risk of prostate cancer compared with those with lower consumption (<2 cups per day) [72]. Another study supporting the potential beneficial effect of coffee consumption is a population-based case-control study reported by Russnes et al where high coffee consumption (>6 cups per day) was associated with reduced risk of high grade (OR 0.45, 95% CI: 0.22-0.90; P < 0.05) and fatal prostate cancer [76].…”

“…There are studies that have investigated the relationship between coffee consumption and risk of prostate cancer [55,[69][70][71][72][73][74][75]. There are those which have found an inverse relationship between coffee consumption and risk of prostate cancer [73][74][75].…”

“…In various cancer entities, positive effects of coffee or caffeine have been described, for instance, in prostate cancer, breast cancer, and skin cancer. In a population cohort study, patients with newly diagnosed prostate cancer had a lower coffee consumption compared to the disease-free population [26]. Subjects with the highest consumption (>3 cups/day) had a 53% lower prostate cancer risk compared to those with the lowest consumption.…”

“…Subjects with the highest consumption (>3 cups/day) had a 53% lower prostate cancer risk compared to those with the lowest consumption. Further investigations in prostate cancer cell lines found that caffeine significantly reduced their proliferative and metastatic behavior [26]. Regarding caffeine and breast cancer, protective effects of caffeine may be mediated through a reduced breast volume.…”
