The inhibitory effect of dehydroepiandrosterone and its derivatives against influenza A virus in vitro and in vivo

Abstract: The antiviral activity of dehydroepiandrosterone (DHEA) and 21 synthetic derivatives against influenza A virus (IAV) replication was investigated in vitro in cell culture. Our results revealed that three DHEA analogues were potent inhibitors of IAV multiplication in MDCK cells and mainly blocked the post-attachment stage of viral infection. Among these derivatives, one containing a 2-OH-Ph moiety (3i) exhibited the best inhibitory effects against H1N1 and H3N2 IAV in a dose-dependent manner. Moreover, treatmen… Show more

“…We predict that TCM can treat diseases through metabolism. DHEA is a key metabolite of cholesterol and affects the inflammatory response of animals (Zhao et al, 2020 ), and has been shown to affect immune function (Yang et al, 2016 ). DHEA appears to treat E. coli ‐induced enteritis in mice through reducing the expression of pro‐inflammatory factors.…”

“…The antiviral properties of steroidal bioconjugates have also been explored and reported. Qingyu Yang et al developed some dehydroepiandrosterone (DHEA) based bioconjugates having potent efficacy against DNA and RNA viruses . Among the several synthesized compounds, DHEA-conjugate 110 bearing a 2-OH-Ph moiety (Figure ) exhibited the best inhibitory effects against H1N1 and H3N2 influenza virus type A (IAV) in a dose-dependent manner.…”

Treatment with conjugate 110 decreased progeny virus yield, viral RNA synthesis, and protein expression.…”

“…Oral DHEA replacement in Addison's disease leads to restoration of levels of regulatory T cells and the decrease in the NK cells (Coles et al 2005). The inhibitory effect of DHEA analogs in viral multiplication has been demonstrated in influenza A virus infection where it inhibited the viral infection in the post-attachment stage (Yang et al 2016).…”
