Shotgun Lipidomic Profiling of the NCI60 Cell Line Panel Using Rapid Evaporative Ionization Mass Spectrometry

Abstract: Rapid Evaporative Ionization Mass Spectrometry (REIMS) was used for the rapid mass spectrometric profiling of cancer cell lines. Spectral reproducibility was assessed for three different cell lines and extent of inter-class differences and intraclass variance were found to allow the identification of these cell lines based on the REIMS data. Subsequently, the NCI60 cell line panel was subjected to REIMS analysis and the resulting dataset was investigated for its distinction of individual cell lines and differe… Show more

“…Other ions at lower m/z values (150–400 m/z) were likely to be derived from fatty acids, rhamnolipids, ceramides and short-chain mycolic acids. These classes of molecules have been identified previously by REIMS/tandem mass spectrometry [19, 20], although there is little prior information about lipid profiles of rodent faeces or the distribution between signals of animal or bacterial origin. The identity of the ions is not critical for these analyses, as it is the pattern of ions that are assessed.…”

“…78 REIMS has also been implemented as an attractive platform for fast shotgun lipidomic profiling of human cancerous cell lines with minimal sample preparation. 104 These striking studies illustrate the capability of ambient-MS in assisting histological pathologists.…”

“…In comparison with MALDI TOF shotgun lipidomics, ambient ionization techniques that do not require matrix such as DESI (desorption electrospray ionization) [129,130] and REIMS (Rapid evaporative ionization mass spectrometry) [131] have recently entered the rather crowded shotgun lipidomic arena, indicating the increasing awareness of the importance of this field. All these approaches target lipid analysis or lipid imaging using the similar ESI method with modification, with the mission that lipid profiling can be completed within a short period of time with the simplest sample preparation to the norm of shotgun lipidomics; rather than the comprehensive analysis conducted by the conventional approach.…”
