TNFα sensitizes neuroblastoma cells to FasL-, cisplatin- and etoposide-induced cell death by NF-κB-mediated expression of Fas

Abstract: BackgroundPatients with high-risk neuroblastoma (NBL) tumors have a high mortality rate. Consequently, there is an urgent need for the development of new treatments for this condition. Targeting death receptor signaling has been proposed as an alternative to standard chemo- and radio-therapies in various tumors. In NBL, this therapeutic strategy has been largely disregarded, possibly because ~50-70% of all human NBLs are characterized by caspase-8 silencing. However, the expression of caspase-8 is detected in … Show more

“…In addition to the mitochondrial apoptotic pathway, ethanol also stimulates the extrinsic apoptotic pathway, which is mediated by death receptors like Fas and the Fas ligand (FasL). 31) Fas and FasL are target genes of nuclear factor-B (NF-κB), 32) and GSTD has been shown to suppress proinflammatory response through blocking the NF-κB pathway. 33) It is possible that GSTD may also inhibit the extrinsic apoptotic pathway in preventing ethanol-induced hepatocellular apoptosis, which merits further investigation.…”

“…Elevated NF-κB activation in cancer cells promotes tumor survival and progression [1921]. In NB, the up-regulation of NF-κB has been shown to accelerate tumor growth and promote cancer cell survival [1, 19, 22, 23].…”

“…Viral myocarditis is a common infectious form of acute or chronic heart muscle inflammation, which can be characterized as either localized or diffuse inflammatory lesions, and is highly related to viral infection (Galenkamp et al, 2015). Generally, more than 5% of individuals with a viral infection will experience some form of myocarditis during the infection period with different clinical manifestations.…”
