Gastrodin Protects against Ethanol-Induced Liver Injury and Apoptosis in HepG2 Cells and Animal Models of Alcoholic Liver Disease

Abstract: This study aims to investigate the protective effects of gastrodin (GSTD), a natural compound isolated from the root of Gastrodia elata BL., on ethanol-induced liver injury and apoptosis in HepG2 cells and animal models. For in vitro studies, GSTD was used to pre-treat the cells for 4 h followed by 600 mM of ethanol co-administration for 24 h. Alcoholic liver disease (ALD) of Sprague-Dawley (SD) rats was induced by chronic ethanol-feeding plus a single dose (5 g/kg) of acute ethanol administration, GSTD at dif… Show more

“…H&E staining was carried out to determine the alterations in the histological features of the liver [ 26 ]. Chronic ingestion of ethanol causes severe pathological damage in the liver, such as necrosis, inflammation, and tissue destruction [ 56 ]. Accordingly, necrosis of the hepatocytes, chromatin condensation, degeneration of the hepatocyte structure, and dilation of the sinusoid, were observed in the ALD model.…”

“…Traditionally, Gas is used on central nervous system disorders such as dementia [36], Alzheimer’s disease [37], Parkinson’s disease [38], affective disorders, and so on. New studies find that Gas can protect against alcohol-induced liver injury in mice (50, 80, 100mg/Kg) and rats (100 mg/Kg) through inhibition of oxidative stress [39,40]. Gas can also inhibit high-glucose-induced human retinal endothelial cell apoptosis by regulating the SIRT1/TLR4/NF-κBp65 signaling pathway [41].…”
