MicroRNA Signatures as Biomarkers and Therapeutic Target for CNS Embryonal Tumors: The Pros and the Cons

Abstract: Embryonal tumors of the central nervous system represent a heterogeneous group of childhood cancers with an unknown pathogenesis; diagnosis, on the basis of histological appearance alone, is controversial and patients’ response to therapy is difficult to predict. They encompass medulloblastoma, atypical teratoid/rhabdoid tumors and a group of primitive neuroectodermal tumors. All are aggressive tumors with the tendency to disseminate throughout the central nervous system. The large amount of genomic and molecu… Show more

“…Anti‐TNF‐α therapy has a profound effect on the serum level of circulating miRNAs in psoriasis; however, these are not related to disease severity . The therapeutic application of miRNAs involves two approaches: miRNA antagonists and miRNA mimics/replacement. miRNA antagonists inhibit endogenous miRNAs.…”

“…The role of miRNAs includes directing and regulating the gene expression of the CSCs. Aberrant expression of miRNA is commonly observed in many types of tumors [31, 32]. One such miRNA, let-7, was found to help control the self-renewal and tumorigenicity in breast, lung, and many other types of cancers [33, 34].…”

“…miRNAs have the ability to regulate the expression of multiple targets instead of a specific target such as small interfering RNA (siRNA) and specific proteins such as BMP . By being able to regulate multiple targets that are involved in a specific signaling cascade with just one molecule, the delivery of miRNA can be advantageous compared to other biomolecules or drug …”
