Cloning and expression analysis of Foxl2 during the reproductive cycle in Korean rockfish, Sebastes schlegeli

Abstract: Foxl2 is a member of the winged helix/forkhead family of transcription factors and is known to regulate ovarian aromatase, which plays a crucial role in ovarian differentiation. To address the role of Foxl2 in gonads and brain during gonadal development, we isolated the full-length cDNA of Foxl2 and analyzed its spatiotemporal expression patterns in the viviparous teleost Korean rockfish, Sebastes schlegeli. Tissue distribution pattern revealed that the Foxl2 was detected in the liver, fat, gill, brain, and ov… Show more

“…Therefore, foxl2 may have a regulatory relationship with cyp19a1a, and the transcription of cyp19a1a gradually increases from August to November and peaked in November. A previous study also revealed that cyp19a1a and cyp19a1b were co-expressed with foxl2, except in the early yolk stage of the ovary [24]. In summary, during fish gonadal development, the highest transcription level of foxl2 was earlier than cyp19a1a, suggesting that foxl2 is involved in fish gonadal differentiation and the maintenance of ovarian function [64].…”

“…Foxl2 is also highly expressed in Nile tilapia XX gonads as early as 5 days after hatching (dah), increases linearly until 35 dah, and remains high at 70 dah (Ijiri et al 2008). However, foxl2b increases from immature stages and peaks at the mature and spawning stage (Figure 10), similar to Foxl2 in chicken (Govoroun et al 2004), Korean rockfish (Mu et al 2013), and African sharptooth catfish (Sridevi and Senthilkumaran 2011). The sequential and divergent expression patterns of zebrafish foxl2a and foxl2b during oogenesis ( Figure 10) indicate that foxl2a and foxl2b may have undergone subfunctionalization after their divergence.…”

“…Differentially expressed genes (DEGs) identified in the transcriptome analysis were chosen and subsequently validated through qRT-PCR. For this purpose, primers were designed using Primer Premier 5.0 software [25], and synthesized by Qingdao Ruibo Biotechnology Co., Ltd., Qingdao, China (Supplementary Material Table S1), primers reference [19,26,27]. The QuantiNova SYBR Green PCR Kit (Qiagen, Dusseldorf, NRW, Germany) was used for the qRT-PCR experiments.…”
