The similarity between human embryonic stem cell-derived epithelial cells and ameloblast-lineage cells

Abstract: This study aimed to compare epithelial cells derived from human embryonic stem cells (hESCs) to human ameloblast-lineage cells (ALCs), as a way to determine their potential use as a cell source for ameloblast regeneration. Induced by various concentrations of bone morphogenetic protein 4 (BMP4), retinoic acid (RA) and lithium chloride (LiCl) for 7 days, hESCs adopted cobble-stone epithelial phenotype (hESC-derived epithelial cells (ES-ECs)) and expressed cytokeratin 14. Compared with ALCs and oral epithelial c… Show more

“…Immunocytochemical studies reveal that HAT-7 cells exhibit several ameloblast characteristics, including the expression of ameloblastin and amelogenin (Kawano et al, 2002). Additionally, expression profiling has shown maturation-stage ameloblast marker proteins such as amelotin and kallikrein-4 (Klk4) are also expressed in these cells (Harada et al, 2006;Matsumoto, Harada, Saito, & Taniguchi, 2011;Yoshizaki et al, 2008;Zheng, Linthicum, DenBesten, & Zhang, 2013).…”

“…Human fetal OEs and ALCs were cultured in order to investigate the function of miR-224 in human epithelial cells in vitro. OEs and ALCs are derived from the same progenitor cell source, but OE is not secretory epithelium (12). In the present study, the miR-224 mimic and inhibitor were transfected into either cultured ALCs or OEs.…”

“…Human fetal oral buccal mucosal epithelial cells (OEs) and ALCs were cultured as described previously (12) in KGM-2 medium (Lonza, Basel, Switzerland) with 1% penicillin/streptomycin, and 0.05 mmol/liter calcium on BD Primaria tissue culture dishes (BD Biosciences, Franklin Lakes, NJ). Clonally selected cells were grown at either pH 7.2 with 0, 10, and 50 M fluoride or at pH 6.2 without fluoride for 72 h.…”

“…Still, only a small percentage of these explants in subrenal cultures demonstrated the formation of dental enamel [60]. Embryonic stem/progenitor cells were similarly demonstrated to differentiate into oral ectoderm and dental epithelium, using variable concentrations of BMP-4 [61]. The formed dental epithelium, when mixed with mouse embryonic dental mesenchyme and transplanted into renal capsules for thirty days, subsequently generated teeth-like structures, including dentin and enamel, with an incisor-like appearance [28].…”
