2-phenylethynesulfonamide inhibits growth of oral squamous cell carcinoma cells by blocking the function of heat shock protein 70

Abstract: Oral squamous cell carcinoma (OSCC) is the most common malignancy in the oral cavity, which accounts for >90% of all diagnosed oral cancers. 2-phenylethynesulfonamide (PES) was known as a selective heat shock protein 70 (Hsp70) function inhibitor, which induced cytotoxic effects on various tumor cell types, but showed to be less toxic to normal cells. However, no associated evaluation of PES on OSCC was found. In the present study, the proliferation of OSCC cells treated with PES was analyzed using a CC… Show more

“…Furthermore, in pleural mesothelioma, VER-155008 inhibits HSP70 by inducing G1 cell cycle arrest and disrupting the PI3K/AKT/mTOR pathway, thereby suppressing tumor proliferation ( Sakai et al, 2021 ). In oral squamous cell carcinoma, PES blocked the cell cycle in the G2/M phase and induced apoptosis ( Jiang and Xiao, 2020 ). In breast cancer, EGCG induced a G2/M phase cell cycle arrest in MCF-7 cells and enhanced the radiosensitivity of breast cancer ( Tran et al, 2010 ).…”

“…SALL4 is a transcription factor containing zinc finger structure, which is specifically expressed in primordial germ cell tumors and plays an important role in early embryonic development. Previous studies (20,21) have shown that the SALL4 initiator is a STAT3-binding region, which can activate cell self-renewal and regulate the function of embryo-fetal stem cells. Its conduction pathway is closely related to the expression of tumor cells, but its specific mechanism and role in tumor metastasis have not been clarified.…”
