Die Azetidinone 1 geben nach Deprotonierung mit LDA bei der Reaktion mit Carbonylverbindungen die substituierten 3-Methylen-~lactame 35. Als Nebenprodukt entstandene Vinylsilane 6 kiinnen auch gezielt aus 1 gewonnen werden 2-Azetldlnones: Reaction of a-Silylcarbanlons with Carbonyl corn. pounds Azetidinones 1 are deprotonated by LDA and reacted with various carbony1 compounds yielding substituted 3-mthylene-2aoetidinom 3-5. As a byproduct in these mctions. the vinylsilane 6 WFS isolated. 6 may te synthesized directly fmm 1 in good yield.In der vorausgegangenen Mitt.') haben wir die Reaktion der a-Silylcarbanionen mit Mesoxaldureester beschrieben. Hier berichten wir uber W i c h e Reaktionen rnit verschiedenen anderen Carbonylverbindungen, wodurch zahlreiche substituierte 3-Methylenderivate zughglich werden.1.4-Diaryl-2-azetidinone werden durch Reformatzky-Reaktion dargestellt*) und mit Chlortrimethylsilan in die a-Silylverbindungen 1 ubefihrt, aus denen die a-Silylcarbanionen 2 mit Lithiumdiisopropylamin bei -78 'C erzeugt werden. Dann erfolgt Reaktion rnit dem Elektrophil, wobei nach Zugabe der jeweiligen Carbonylverbindung das Kiihlbad entfemt wird, so d d sich die Reaktionsansatze bis auf Raumtemp. erwhnen konnen.Mit aliphatischen Aldehyden erhiilt man 3-Alkyliden-P lactame 3 in einem E/Z-Verh<nis von 1:1, wobei die Diastereomeren sc sauber getrennt werden konnen. Die stereochemische Zuordnung kOnnte durch Vergleich der 'H-NMR-Spektren mit bekannten Daten gemffen werden3). Das Signal fur das Proton an C-4 findet man bei den E-Isomeren stets um 6 = 5.4 ppm, bei allen Z-Isomeren bei 6 = 5.25 ppm; das exocyclische a-Proton der E-Isomere tritt bei 6 = 6.1-6.3 ppm, das der Z-Isomere bei 6 = 5.4-5.6 ppm in Resonanz. Die Kopplung zwischen 4-H und a-H ist unabhhgig von der Konfguration bei allen Beispielen J = 1.5 Hz.Um die acceptorsubstituierten Verbindungen 4 zu erhalten, wurde 2 mit a-Ketoestem zur Reaktion gebracht. So erhielten wir rnit Phenylglyoxylsauremethylester 4a und 4b, mit Brenztraubensauremethylester 4c,d und mit Brenztraubensaureethylester 4e,f. Diese Verbindungen entstehen in einem E/ZVerhiiltnis 1 : 1 und die Isomeren konnten sc getrennt und isoliert werden. Die Strukturauklhng erfolgte rnit Hilfe von NOE-IJntersuchungen.Setzten wir 2 mit Formamiden um, so beobachteten wir eine analoge Reaktion, die zur Bildung der Enamin-fL carbonsaureamide 5 fiihrt. Derartig substituierte PLactame sind auf anderem Wege, durch Photolyse von 4Pyrimido-nen4), bereits dargestellt worden. An anderen a-lithiierten Silylverbindungen konnte mit Amiden eine analoge Umsetzung beobachtet werden'). Durch NOE-Untersuchungen konnten wir feststellen, dal3 die von uns isolierfen Verbindungen stets die E-Konfiguration aufweisen. Spektroskopische Untersuchungen der Rohprodukte ergaben jedoch, das auch hier beide Diasteremomere entstehen. Die E-Form durfte thermodynamisch begunstigt sein, da bereits nach kurzem Erhitzen der Anteil an E-Isomeren zunimmt, und es war uns weder sc noch durch Umkristallisation moglich, die entsprechende ...

Dicarboxylates of 3‐Methylidene‐β‐lactams: Addition Reactions to the Exocyclic Double Bond, Formation of Spiro‐β‐lactams, and Reductive Ring Opening by Hydrazines

Horst Bohme, Marburg, on the occasion of his 85th birthday (1 6.V11.93) 0-, S-, and N-Nucleophiles are added to the exocyclic double bond of the title compounds 1. The addition of 0-or S-nucleophiles yields stable products (Scheme I ) , while addition of N-nucleophiles results in thermally labile compounds (Scheme 2). The reaction is studied by spectroscopic methods. From hydrazine adducts, a spiro[azetidine-3,3'-pyrazolidine] 7 is obtained, and the addition products of methyl-and benzylhydrazine rearrange to pyrazo~-4-cdrboxylates 6. Furthermore, the exocyclic double bond is used for the formation of spiro-p-lactams either by cyclopropane formation or by Diels-Alder reactions (Scheme 4 ) . The steric course of all reactions is studied, and it is shown that all reactions with the double bond occur from the side opposite to the bulkier substituent at C(4) of the p-lactam ring.Introduction. -Since the discovery of p-lactam antibiotics with an exocyclic double bond at C(7) like asparenomycins [I], the interest in other similar structures, e.g. monocyclic p-lactams with this structural component has grown [2]. Nucleophiles, e.g. amines [3] or thiols [4], are added to the double bond of unsubstituted 3-methylidene-P-lactams and do not open the lactam ring. Therefore, it seems to be of special interest to study the behaviour of substituted analogues. Principially, two types of substitution may be differentiated: 1) alkyl or aryl groups connected to the double bond directing a nucleophilic attack to the exocyclic C ( a ) atom or 2) electron-withdrawing groups at this position directing the attack towards C(3) of the p-lactam ring. In preceding papers 151, we described the synthesis of compounds 1 from 3-silylated /I-lactams and mesoxalic esters as model compounds of the second type of substitution. Now we wish to present some reactions at the double bond of 1 which show the synthetic possibilities and might allow a look into their mode of action when used in biological systems. These results should give some hints onto the hitherto unknown mechanism of the action of methylidene-P-lactams as 8-lactamase inhibitors.

Reaction of the 3-silylated -1actams 1 with glycoxalates gives bis-lactam 3, but the same reaction in the presence of 1 equiv. of Me,SiC1 leads to the formation of the disilylated adducts 5. The latter is desilylated by (Bu,N)F yielding the monocarboxylates 7 of 3-methylideneB -lactams, which, with oxidizing agents, give the spiro compound 8. The structure of 8 is established by spectroscopic data and a crystal-structure analysis.Introduction. -As described in a previous paper [l], the epoxidation of the electronpoor double bond of dicarboxylates of 3-methylidene4 -1actams can be effected by a large variety of oxidizing agents yielding spiro[azetidine-3,2'-oxirane] derivatives. But until now, all experiments to open the oxirane ring by nucleophiles failed. Probably, a nucleophilic attack is not possible at the C-atoms of the tetrasubstituted oxirane. A nucleophilic ring opening of the epoxide would afford an easy route to introduce a heteroatom either into position 3 of thea-lactam ring or into the a-position of the side chain, and thereby open the possibility to develop an alternative route to potent antibiotics, hopefully being more stable againstp -1actamases [2]. Therefore, we tried to synthesize the monocarboxylates 7 of 3-methylidene4 -1actams and the corresponding trisubstituted epoxides.
