Cobalt(II) and nickel(II) complexes with diclofenac (dicl) in the presence of nitrogen-donor 3-picoline (3-pic) and 1-(2-aminoethyl)pyrrolidine (2-aepyr), [Co(dicl) 2 (3-pic) 2 ] (1), [Ni(dicl) 2 (3-pic) 2 (H 2 O) 2 ] (2), [Co(dicl) 2 (2-aepyr) 2 ] (3), and [Ni(dicl) 2 (2-aepyr) 2 ] (4), have been synthesized and characterized by FT-IR, UV-Vis, elemental and thermal analysis. The crystal structures of 1 and 3 have been determined by single-crystal X-ray diffraction; phase purity of complexes has been proved by powder X-ray diffraction analysis. Structural analyses have demonstrated that 1 and 3 are mononuclear and Co(II) ions have distorted octahedral environments. In 1, dicl is bidentate, whereas in 2, 3, and 4 dicl is monodentate. Dicl ligands are coordinated to metal(II) ions with O of carboxylate. Therefore, IR spectra of all complexes display {ν(OCO) asym and ν(OCO) sym } of dicl. The calculated Δν(OCO) values are consistent with the presence of monodentate (>200) and bidentate (<200) carboxylate. Compounds 2, 3, and 4 exhibit inhibition effects on human carbonic anhydrase-I. Compounds 3 and 4 show high thermal stability compared with 1 and 2.

The conversion of carbon dioxide (CO ) and bicarbonate (HCO ) to each other is very important for living metabolism. Carbonic anhydrase (CA, E.C.4.2.1.1), a metalloenzyme familly, catalyzes the interconversion of these ions (CO and HCO ) and are very common in living organisms. In this study, a series of novel 2-amino-3-cyanopyridines supported with some functional groups was synthesized and tested as potential inhibition effects against both cytosolic human CA I and II isoenzymes (hCA I and II) using by Sepharose-4B-l-tyrosine-sulfanilamide affinity chromatography. The structural elucidations of novel 2-amino-3-cyanopyridines were achieved by NMR, IR, and elemental analyses. K values of the novel synthesized compounds were found in range of 2.84-112.44 μM against hCA I and 2.56-31.17 μM against hCA II isoenzyme. While compound 7d showed the best inhibition activity against hCA I (K : 2.84 μM), the compound 7b demonstrated the best inhibition profile against hCA II isoenzyme (K : 2.56 μM).

Effects of clindamycin, dexketoprofen trometamol, benzylpenicillin (penicillin G) and phenoxymethylpenicillin (penicillin V) were examined on purified sheep carbonic anhydrase I and II (SCAI, SCA-II). In our study, we were chosen sheep liver tissue; because the liver is responsible for detoxification of xenobiotics. The CA isozyme have a very important duties on organisms, the effect of xenobiotics on the CA enzyme was reported. It was observed these drugs exhibited the inhibition effects against liver carbonic anhydrase isozymes I and II in our study. SCA I and SCA II isozymes were purified with about a specific activity of 1108.51 EU/mg and 5706.67 EU/mg, 120.62 and 620.96 purification fold by using Sepharose-4B-L tyrosine-sulfanilamide affinity gel chromatography. Purity of the SCA I and SCA II isozyme was checked by SDS-PAGE technique and subunit molecular masses of the SCA I was found as 29 kDa and SCA II found as 31 kDa. The usage of clindamycin, dexketoprofen trometamol, benzylpenicillin (penicillin G) and phenoxymethylpenicillin (penicillin V) in sheep liver especially during necrotic hepatitis disease is important for scientific researches. Many antibiotic and antihistamine may have dangerous effect to sheep liver. Also, it is important to study the inhibition effects of these drugs on SCA I and II isozymes.
