17-β estradiol in the acetylcholinesterase activity and lipid peroxidation in the brain and blood of ovariectomized adult and middle-aged rats

Abstract: 17-β estradiol is able to modulate the AChE activity and non-neuronal cholinergic response as well as to reduce lipid peroxidation. Its response is dependent on the age and brain structure analyzed.

“…A selective ER-b agonist more markedly reduced (52.5%) this activity toward reference levels in Ovx-Dia rats than did E 2 (40.2%) and an ER-a agonist (21.1%). These results corroborate previous studies which found that treatment with estradiol decreases acetylcholinesterase activity in the brain of ovariectomized 46 and diabetic 48 rats. A study by Huang and Zhang 49 reported that 7-O-modified genistein, a selective ER-b agonist, showed anticholinesterase activity in ovariectomized rats.…”

“…47 Acetylcholinesterase, a surrogate marker of dementia, is increased after menopause and diabetes. 46,48 We also observed a significant increase in acetylcholinesterase activity (58.2%) in Ovx-Dia rats compared with their respective controls. A selective ER-b agonist more markedly reduced (52.5%) this activity toward reference levels in Ovx-Dia rats than did E 2 (40.2%) and an ER-a agonist (21.1%).…”

“…Measure of the AChE activity can provide an evidence, at least indirectly, of how the concentration of Ach is in the brain [30]. One recent research result showed that AChE in striatum of rats that received bilateral ovariectomy was activated [31]. Our results demonstrated that the activity and expression of AChE in hippocampus of OVX animals were both increased compared with control animals.…”

“…During ageing (Rastogi et al 2012) and ovariectomy (Martins et al 2012), the enhanced generation of ROS induced chronic oxidative stress in the brain. This condition caused gradual accumulation of damaged proteins leading to a functional decline in the brain's endogenous defence system (Rumora et al 2007;Min et al 2008) and leading to the activation of stress kinases (JNK and p38 MAPK) (Gabai et al 1997).…”
