<b>Population structure and impact of recurrent selection on popcorn using EST-SSR markers

Abstract: The success of any recurrent selection program depends on the genetic variability of the evaluated population, which is used to refer to the diversity of existing alleles at many genetic loci. Thus, the goal of the present study was to investigate the impact of recurrent selection across nine cycles of a UENF-14 popcorn population through the analysis of genetic diversity and structure using microsatellite markers (EST-SSRs). Genomic DNA was extracted from young leaves of 25 individuals from each cycle (C0, C1… Show more

“…The amplified product size was separated by gel electrophoresis in 4% high-resolution agarose (Guimaraes et al 2018) gel stained in ethidium bromide solution in 1× TAE buffer at a constant 80 V current for 4 h. To visualize the bands, gel was observed in UV light provided by a trans-illuminator in (Syngene G: Box) gel documentation system. The size and molecular weight of the amplification products was estimated by a standard 50 bp ladder (Fermentas Thermofisher Pvt.…”

“…Since its inclusion in the germplasm bank of the State University of Northern Rio de Janeiro (UENF), the UENF-14 popcorn population has been subject to a series of recurrent intrapopulational selection cycles targeting the improvement of economically important traits and the maintenance of the population variability. Until the eighth cycle, only phenotypic evaluations were performed [1220]. The population is currently in the ninth selection cycle (C-9), and the introduction of genotyping proved important for predicting gains for the main traits of interest as well as for allowing a more detailed study of the genomic regions responsible for such traits.…”

“…These results indicate that even with the use of different progeny strategies (full-sibs, S 1 and halfsibs), there was an increase in allele frequencies for the traits of interest, with greater adaptability to counties in the north and northwest of the State of Rio de Janeiro. This progress has been described by Ribeiro et al (2016) and Guimarães et al (2018b). While the first authors investigated cumulative gains in GY and PE based on phenotypic responses in the same study population over seven cycles (C0-C6), the latter authors used microsatellite molecular markers (EST-SSRs), inferring an increase in favourable alleles over nine selection cycles (C0-C8), which may be related to the main traits of interest.…”
