The present study was aimed to determine the effect of ATP dose rate on longevity, motility, and viability of frozen-thawed buffalo bull’s sperm. For this purpose, 20 frozen semen straws of four different buffalo bulls were obtained from a private semen production unit (Al-Hiawan Sires, Sahiwal). The experiment was replicated four times. In each replicate, four frozen straws of the same bulls were thawed and pooled. After evaluating the motility, the pooled frozen-thawed sample was aliquoted into three separate tubes containing 0 (control), 1-, or 2-mM concentrations of ATP. Afterward, each aliquot was incubated at 37oC for 4 hrs. The sperm motility, viability, and membrane integrity were observed at various time points (10 min., 1hr, 2hr, and 4hr) during the incubation period of four hrs. It was observed that progressive motility and most of the motion kinematic parameters were greater (P<0.05) in control. A significant (P<0.05) interaction was observed between ATP dose and incubation time. The progressive motility, total motility, VCL, VAP, ALH, and BCF decreased (P<0.05) both by incorporation of ATP (1mM or 2mM) and increasing incubation time (from 10 min to 4hrs) which indicate the ATP and incubation time of 4 hours negatively affect the motion kinetics of buffalo bull sperm. In conclusion, it was witnessed that the motility and plasma membrane integrity of sperm declined by the supplementation of ATP in frozen-thawed buffalo bull semen.

Technologies Used in Production Systems for Santa Inês Sheep: A Systematic Review

This study identifies the number of publications that presented technologies used in the production systems of Santa Inês sheep in the last 5 years (2017–2021) carried out in Brazil. Therefore, the objective was to identify where we are in terms of knowledge about technologies in different fields (health, reproduction, animal breeding, behavior and welfare, nutrition and feeding, forage and pasture, carcass and meat quality, and economics and management of livestock systems). After rigorous selection, 114 studies were appointed and classified by knowledge field, and the main approaches within each theme were evaluated, pointing out research gaps. Most technologies have been in northeastern states. However, government agencies should develop public policies to disseminate techniques in rural areas because the production system in this region is still subsistence. This study highlighted the need for works that present management practices and tools that impact the improvement of animal welfare. Agro-industrial by-products have been widely used as an alternative for sheep feeding. However, economic feasibility analyses are recommended with these foodstuffs to substantiate their use as an option to reduce production costs. There is a lack of research allusive to the management of production systems, especially those related to estimates of economic feasibility indicators.

The oxidative stress constitutes accumulation of free radicals (FR) in the mammalian cells and tissue. Elevated accumulation of reactive oxygen species (subset of free radicals that contain oxygen) lead to a number of pathological abnormalities in cellular functioning respective to cells and organ damage. Excessive reactive oxygen species (ROS) formation can induce significant changes in sperm morphology, vitality, and oocyte fertizability. Spermatozoa membrane is composed of polyunsaturated fatty acids, which undergo oxidation under the action of free radicals. These processes result in reduced motility and impaired ability of sperm to fertilize the ovum. The antioxidants are the natural inhibitors of ROS activity. In the semen, they are found in large quantities in the seminal plasma. Their biological role is to attenuate the damaging effects of ROS. Low amounts of ROS are involved in the normal physiological and metabolic processes of the animal organism. Various reasons related to rearing, feeding and exploitation of the male animals can cause their overproduction. The imbalance between natural antioxidants and ROS production and accumulation result into body falling into the so-called oxidative stress (OS). This effect negative on various cell organelles. Damage to the sperm plasma membrane is caused by an imbalance between reactive oxygen species and antioxidants, which occurs especially during the semen processing (dilution, freeze-thaw). The aim of this literature review is to summarize the mechanism of influence of the oxidative stress and the possibilities to reduce its harmful effect over the ram semen parameters after chilling or freezing process.
