Screening of mucoadhesive vaginal gel formulations

Abstract: Rational design of vaginal drug delivery formulations requires special attention to vehicle properties that optimize vaginal coating and retention. The aim of the present work was to perform a screening of mucoadhesive vaginal gels formulated with carbomer or carrageenan in binary combination with a second polymer (carbomer, guar or xanthan gum). The gels were characterised using in vitro adhesion, spreadability and leakage potential studies, as well as rheological measurements (stress and frequency sweep test… Show more

“…Table I. Based on results of a previous work, in which hydrogels developed using the same polymers were characterized using rheological and spreadability measurements [19], the combination of Cg and Gg was not included in the present work. Hydrogels 1 and 5 (CbPe0.5 and CbXg1.5) were also formulated including 2% of caffeine to perform active release studies.…”

“…Vaginal dosage forms have a short residence time at the place where the drug is applied, therefore, for its control as well as determination of fixation degree the rates of distribution and adhesion have been determined [7,8].…”

“…This indicates the force of the interaction between hydrogel and the mucin. ∆F max values of formulations containing Satiaxane ® , F8, F9, F15 and F16, were higher than those without [45], ophthalmic hydrogel [46] or vaginal hydrogel [47]. As a natural biocompatible polymer, xanthan gum has a β-(1→4)-D-glucose backbone where every second glucose unit is attached to a trisaccharide consisting of mannose, glucuronic acid, and mannose.…”
