Detection and molecular characterization of cactus witches'-broom disease associated with a group 16SrII phytoplasma in northern areas of China

Abstract: In July 2010, cacti with typical phytoplasma symptoms were observed in Yangling district, Shaanxi Province, China. Based on amplification of 16S rRNA gene, phytoplasmas were detected by polymerase chain reaction (PCR) driven with universal primer pairs P1/ P7, followed by a nested PCR with universal primer pairs R16F2n⁄R16R2. Fragments of expected sizes (1.8 kb and 1.2 kb) were obtained from symptomatic samples, but not from the asymptomatic samples. Sequencing results and BLASTn analysis of the 1806 bp produc… Show more

“…The reason of the unspecific digestion was kept unknown. It is known that phytoplasmas of 16SrI are the main group occurred in China (Li, Zhang, Liu, et al, 2012), and various subgroups and novel subgroups were discovered (Li, Zhang, Liu, et al, 2012; Li et al., 2014; Luan et al., 2018; Tseng et al., 2016; Zhang et al., 2019). In this work, we extended the queue of 16SrI subgroups with another two novel subgroups of 16SrI‐AK and ‐AL.…”

“…Groups 16SrI and ‐V are important in northern areas of China, and group 16SrII is important in southern areas of China (Li et al. ). This, however, is the first record of group 16SrVI phytoplasma in China, which, to our knowledge, has great significance for plant pathology research in China.…”

“…In 2009, locust trees witches' -broom disease was found in Taian, Shandong (Yu et al, 2012). In July 2010, the cactus witches' -broom disease caused by the 16SrII-C subgroup was first reported in Yangling, Shanxi (Li et al, 2012). Tomato yellowing disease caused by a 16SrII-A phytoplasma was first reported in Yunnan Province in 2011 (Dong et al, 2013).…”
